The supply of electrical energy is critical to convenient and comfortable living. However, people consume a large amount of energy, contributing to an energy crisis and global warming, and damaging some ecological cycles. Residential electricity consumption has greater elasticity than industrial and business consumption; it therefore has high energy-saving potential. This work establishes an automated platform, which provides information about residential electricity consumption in each city in Taiwan. Machine learning was used to forecast future residential electricity demand. A nature-inspired optimization method was applied to enhance the accuracy of the best machine learner, yielding an even better hybrid ensemble model. Performance measures indicate that the resulting model is accurate and provides effective information for reference. An automatic web-based system based on the model was combined with a web crawler and scheduled to run automatically to provide information on monthly residential electricity consumption in each county and city. By providing energy consumption information across the country, power providers and government can discuss policy and set different goals for energy use. The results of this study can facilitate the early implementation of energy-saving and carbon emission-reducing in cities and aid utility companies in establishing energy conservation guidelines.  相似文献

Chrysanthemum flower is among one of the highly sought after and widely planted flower crops, in particular for cultural and religious ceremonies. However, the chrysanthemum stem and stalk have little value and usually discard as by‐product waste from floristry. The objective of this research is to investigate the potential value of utilizing chrysanthemum stem and stalk as reinforcing fillers for thermoplastic composites. In this study, 2‐mm thick composite sheet containing predefined formulations of polylactic acid (PLA), chrysanthemum waste filler (CWF) ranging from 15 to 60 phr, and maleated polyethylene (MAPE) coupling agent up to 5 phr were prepared with the aid of Haake internal mixer and compression molding. The effect of MAPE loading on tensile, thermal, and morphological properties of PLA/CWF composites was investigated. The findings revealed that PLA/CWF composite attained improved tensile modulus compared to the neat PLA, and the tensile modulus increases with higher concentration of CWF. However, both tensile strength and elongation at break reduces with increase loading of CWF. Overall, PLA/CWF composites with MAPE shows better performance compared to those without MAPE, where an optimum strength of 21.8 MPa can be achieved with 60 phr CW and 3 phr MAPE. Abstract : A major challenge of targeted cancer therapy is the selection for drug-resistant mutations in tumor cells leading to loss of treatment effectiveness. p97/VCP is central regulator of protein homeostasis and a promising anticancer target because of its vital role in cell growth and survival. One ATP-competitive p97 inhibitor, CB-5083, has entered clinical trials. Selective pressure on HCT116 cells dosed with CB-5083 identified five different resistant mutants. Identification of p97 inhibitors with different mechanisms of action would offer the potential to overcome this class of resistance mutations. Our results demonstrate that two CB-5083 resistant p97 mutants, N660 K and T688 A, were also resistant to several other ATP-competitive p97 inhibitors, whereas inhibition by two allosteric p97 inhibitors NMS-873 and UPCDC-30245 were unaffected by these mutations. We also established a CB-5083 resistant cell line that harbors a new p97 double mutation (D649 A/T688 A). While CB-5083, NMS-873, and UPCDC-30245 all effectively inhibited proliferation of the parental HCT116 cell line, NMS-873 and UPCDC-30245 were 30-fold more potent in inhibiting the CB-5083 resistant D649 A/T688 A double mutant than CB-5083. Our results suggest that allosteric p97 inhibitors are promising alternatives when resistance to ATP-competitive p97 inhibitors arises during anticancer treatment.  相似文献

The generation of ammonia, hydrogen production, and nitrogen purification are considered as energy intensive processes accompanied with large amounts of CO2 emission. An electrochemical method assisted by photoenergy is widely utilized for the chemical energy conversion. In this work, earth‐abundant iron pyrite (FeS2) nanocrystals grown on carbon fiber paper (FeS2/CFP) are found to be an electrochemical and photoactive catalyst for nitrogen reduction reaction under ambient temperature and pressure. The electrochemical results reveal that FeS2/CFP achieves a high Faradaic efficiency (FE) of ≈14.14% and NH3 yield rate of ≈0.096 µg min?1 at ?0.6 V versus RHE electrode in 0.25 m LiClO4. During the electrochemical catalytic reaction, the crystal structure of FeS2/CFP remains in the cubic pyrite phase, as analyzed by in situ X‐ray diffraction measurements. With near‐infrared laser irradiation (808 nm), the NH3 yield rate of the FeS2/CFP catalyst can be slightly improved to 0.1 µg min?1 with high FE of 14.57%. Furthermore, density functional theory calculations demonstrate that the N2 molecule has strong chemical adsorption energy on the iron atom of FeS2. Overall, iron pyrite‐based materials have proven to be a potential electrocatalyst with photoactive behavior for ammonia production in practical applications.  相似文献

The late-onset type of Fabry disease (FD) with GLA IVS4 + 919G > A mutation has been shown to lead to cardiovascular dysfunctions. In order to eliminate variations in other aspects of the genetic background, we established the isogenic control of induced pluripotent stem cells (iPSCs) for the identification of the pathogenetic factors for FD phenotypes through CRISPR/Cas9 genomic editing. We adopted droplet digital PCR (ddPCR) to efficiently capture mutational events, thus enabling isolation of the corrected FD from FD-iPSCs. Both of these exhibited the characteristics of pluripotency and phenotypic plasticity, and they can be differentiated into endothelial cells (ECs). We demonstrated the phenotypic abnormalities in FD iPSC-derived ECs (FD-ECs), including intracellular Gb3 accumulation, autophagic flux impairment, and reactive oxygen species (ROS) production, and these abnormalities were rescued in isogenic control iPSC-derived ECs (corrected FD-ECs). Microarray profiling revealed that corrected FD-derived endothelial cells reversed the enrichment of genes in the pro-inflammatory pathway and validated the downregulation of NF-κB and the MAPK signaling pathway. Our findings highlighted the critical role of ECs in FD-associated vascular dysfunctions by establishing a reliable isogenic control and providing information on potential cellular targets to reduce the morbidity and mortality of FD patients with vascular complications.  相似文献

Fully dense and magnetically controllable glass ceramics was successfully synthesized by method of hot pressing using CaO-Al2O3-SiO2-Na2O glass powder and Fe3O4 powder as raw materials. The influences of sintering temperature and time, content and particle size of Fe3O4, as well as particle size of glass powder on the densification and magnetic properties of samples were investigated. It was found that the saturation magnetization gradually increased with increasing magnetite content. In addition, the samples containing smaller size magnetite particles had a higher coercivity. However, for samples using smaller size glass powder, magnetite particles could partially dissolve into the glass matrix, which led to the decrease of saturation magnetization and the increase of coercivity. It was also concluded that the precipitation of crystalline phase from smaller size glass powder caused the decrease of degree of densification, and after decreasing the sintering temperature, the degree of densification of product was enhanced.  相似文献
