Leucine-rich repeats containing G protein-coupled receptor 4 (LGR4) is a receptor that belongs to the superfamily of G protein-coupled receptors that can be activated by R-spondins (RSPOs), Norrin, circLGR4, and the ligand of the receptor activator of nuclear factor kappa-B (RANKL) ligands to regulate signaling pathways in normal and pathological processes. LGR4 is widely expressed in different tissues where it has multiple functions such as tissue development and maintenance. LGR4 mainly acts through the Wnt/β-catenin pathway to regulate proliferation, survival, and differentiation. In cancer, LGR4 participates in tumor progression, invasion, and metastasis. Furthermore, recent evidence reveals that LGR4 is essential for the regulation of the cancer stem cell population by controlling self-renewal and regulating stem cell properties. Methamphetamine is, worldwide, one of the most consumed drugs of abuse. One important side effect is neurodegeneration leading to a decrease in life expectancy. The aim of this paper was to check whether the drug affects one of the receptors involved in neurodegeneration/neuroprotection events, namely the adenosine A2A receptor (A2AR). First, we noticed that methamphetamine does not affect A2A functionality if the receptor is expressed in a heterologous system. However, A2AR becomes sensitive to the drug upon complexes formation with the cannabinoid CB1 receptor (CB1R) and the sigma 1 receptor (σ1R). Signaling via both adenosine A2AR and cannabinoid CB1R was affected by methamphetamine in cells co-expressing the two receptors. In striatal primary cultures, the A2AR–CB1R heteromer complex was detected and methamphetamine not only altered its expression but completely blocked the A2AR- and the CB1R-mediated activation of the mitogen activated protein kinase (MAPK) pathway. In conclusion, methamphetamine, with the participation of σ1R, alters the expression and function of two interacting receptors, A2AR, which is a therapeutic target for neuroprotection, and CB1R, which is the most abundant G protein-coupled receptor (GPCR) in the brain. 相似文献
The volume of tailings produced by the extractive industry has been increasing due to the processing of the low‐grade ore. This issue can cause environmental accidents and require significant investment to control the disposal of tailings. Therefore, this study aims to recover iron from zinc mine tailings by wet magnetic separation followed by the carbothermal reduction of self‐reducing briquettes. Two magnetic separation routes were investigated to concentrate iron. Zinc mine tailings were processed by route I, in a rougher stage followed by a scavenger stage; and route II, in a rougher stage followed by a cleaner stage. The carbothermal reductions were performed using self‐reducing briquettes composed of Fe concentrate from the route with high Fe content and charcoal. The products were analyzed by scanning electron microscopy with energy dispersive spectroscopy (SEM‐EDS), x‐ray diffraction (XRD), inductively coupled plasma optical emission spectrometry (ICP‐OES), and volumetric chemical analysis. Magnetic separation route II provided the highest‐grade Fe concentrate, 52% Fe, while route I provided 33% Fe. In the carbothermal reductions, a metallization degree of 98% in the Fe concentrate briquette, 97% in the briquette with a 10% replacement of its raw material by Fe concentrate, and 99% in the hematite briquette was reached. The replacement of raw material by Fe concentrate showed no significant change in Fe recovery. Considering the whole process, magnetic separation and carbothermal reduction, the recovery of Fe from the zinc mine tailings was 67%. Therefore, the process route suggested in this study will not only reduce tailings disposal and consequently the risk of environmental accidents, but it will also provide profitable raw material for the steel industry. 相似文献
A recent development in tactile technology enables an improvement in the appreciation of the visual arts for people with visual impairment (PVI). The tactile sense, in conjunction with, or a possibly as an alternative to, the auditory sense, would allow PVIs to approach artwork in a more self‐driven and engaging way that would be difficult to achieve with just an auditory stimulus. Tactile colour pictograms (TCPs), which are raised geometric patterns, are ideographic characters that are designed to enable PVIs to identify colours and interpret information by touch. In this article, three TCPs are introduced to code colours in the Munsell colour system. Each colour pattern consists of a basic cell size of 10 mm × 10 mm to represent the patterns consistently in terms of regular shape. Each TCP consists of basic geometric patterns that are combined to create primary, secondary, and tertiary colour pictograms of shapes indicating colour hue, intensity and lightness. Each TCP represents 29 colours including six hues; they were then further expanded to represent 53 colours. Two of them did not increase the cell size, the other increased the cell size 1.5 times for some colours, such as yellow‐orange, yellow, blue, and blue‐purple. Our proposed TCPs use a slightly larger cell size compared to most tactile patterns currently used to indicate colour, but code for more colours. With user experience and identification tests, conducted with 23 visually impaired adults, the effectiveness of the TCPs suggests that they were helpful for the participants. 相似文献
Over the last decade, AMD waters have gained more attention as a potential source of metals due to the emerging need to recover or recycle metals from secondary resources. Metals recovery supports sustainability and the development of a circular economy with benefits for resource conservation and the environment. In this study, five extractants (Acorga M5640, LIX 54, LIX 622, LIX 622 N, and LIX 864) diluted (15% (v/v)) in Shell GTL with 2.5% (v/v) octanol were compared and evaluated for Cu recovery from an extreme AMD sample (5.3?±?0.3 g/L Cu) collected at the inactive São Domingos Mine in the Iberian Pyrite Belt of Portugal. Of the five extractants, Acorga M5640 showed the best selective efficiency. Further tests showed that 30% (v/v) of this extractant was able to selectively extract ≈ 96.0% of the Cu from the AMD in one extraction step and all of the remaining Cu (to below detection) in three steps. Among the different stripping agents tested, 2 M sulfuric acid was the most efficient, with ≈ 99% of the Cu stripped, and the recyclability of the organic phase was confirmed in five successive cycles of extraction and stripping. Furthermore, contact time tests revealed that the extraction kinetics allows the transfer of ≈ 97% of the Cu in 15 min, and aqueous to organic phase ratios tests demonstrated a maximum loading capacity of ≈ 16 g/L Cu in the organic phase. Raising the concentration of Cu in the stripping solution (2 M sulfuric acid) to ≈ 46 g/L through successive striping steps showed the potential to recover elemental Cu using traditional electrowinning. Finally, a biological approach for Cu recovery from the stripping solution was evaluated by adding the supernatant of a sulfate-reducing bacteria culture to make different molar ratios of biogenic sulfide to copper; ratios over 1.75 resulted in precipitation of more than 95% of the Cu as covellite nanoparticles.
A simplified low-cost approach to experimentally determine transport parameters in mountain rivers is described, with an emphasis on the longitudinal dispersion coefficient (DL). The approach is based on a slug injection of table salt (NaCl) as a tracer and specific conductance readings at different locations downstream of the injection spot. Observed specific conductance readings are fit using the advection-dispersion equation with OTIS-P, yielding estimates of cross-sectional area and longitudinal dispersion coefficient for various stream reaches. Estimates of the DL are used to assess the accuracy of several empirical equations reported in the literature. This allowed the determination of complementary transport parameters related to transient storage zones. The empirical equations yielded rather high DL values, with some reaching up an order of magnitude higher to those obtained from tracer additions and OTIS-P. Overall, the proposed approach seems reliable and pertinent for river reaches of ca. 150 m in length. 相似文献
Association between poly(vinyl alcohol-co-vinyl acetate) copolymer (PVA) and sodium dodecyl sulfate (SDS) was studied experimentally and theoretically. It was found that, for the ethyl acetate-aqueous phase interface in which PVA was previously adsorbed, the interfacial tension (γ) increases abruptly to a maximum and then exhibits a relatively mild decay with the addition of SDS to the aqueous phase. The theoretical results indicate that vinyl acetate (VAc) segments determine γ. However, for relatively low concentrations of SDS (CSDS), this latter plays a major role because through its association with the VAc segments it modulates the extent to which PVA is adsorbed at the interface, indirectly determining the value of γ. As CSDS approaches to the CMC value for SDS, its influence on γ decreases because SDS tends to self-assembly rather than associates with VAc. These model predictions are consistent with experimental findings reported in the literature. 相似文献
Probabilistic topic modeling algorithms like Latent Dirichlet Allocation (LDA) have become powerful tools for the analysis of large collections of documents (such as papers, projects, or funding applications) in science, technology an innovation (STI) policy design and monitoring. However, selecting an appropriate and stable topic model for a specific application (by adjusting the hyperparameters of the algorithm) is not a trivial problem. Common validation metrics like coherence or perplexity, which are focused on the quality of topics, are not a good fit in applications where the quality of the document similarity relations inferred from the topic model is especially relevant. Relying on graph analysis techniques, the aim of our work is to state a new methodology for the selection of hyperparameters which is specifically oriented to optimize the similarity metrics emanating from the topic model. In order to do this, we propose two graph metrics: the first measures the variability of the similarity graphs that result from different runs of the algorithm for a fixed value of the hyperparameters, while the second metric measures the alignment between the graph derived from the LDA model and another obtained using metadata available for the corresponding corpus. Through experiments on various corpora related to STI, it is shown that the proposed metrics provide relevant indicators to select the number of topics and build persistent topic models that are consistent with the metadata. Their use, which can be extended to other topic models beyond LDA, could facilitate the systematic adoption of this kind of techniques in STI policy analysis and design.
In this work, biocomposites made of polyhydroxyalkanoates (PHA) with natural fibers were produced via compression molding. In particular, polyhydroxybutyrate (PHB) and polyhydroxybutyrate-co-hydroxyvalerate (PHBV) were reinforced with 20 wt% of agave fibers. Different compatibilization strategies were investigated to improve the fiber-matrix interaction: fiber surface treatment in PHA solution, fiber surface treatment in maleated PHA solution, fiber propionylation, and extrusion with maleated PHA. The biocomposites were characterized in terms of morphology, mechanical properties, water absorption, and biodegradability by CO2 production tracking. In general, fiber propionylation was the best strategy for mechanical properties enhancement and water uptake decreasing. Biocomposites with propionylated fibers showed improved flexural strength (170% for PHB and 84% for PHBV). The flexural modulus was also enhanced with propionylated fibers up to 19% and 18% compared to uncompatibilized biocomposites (PHB and PHBV, respectively). Tensile strength increased by 16% (PHB) and 14% (PHBV), and the water absorption was reduced using propionylated fibers going from 6.6% to 4.4% compared with biocomposites with untreated fibers. Most importantly, the impact strength was also improved for all biocomposites by up to 96% compared with the neat PHA matrices. Our aim was to investigate the subset distribution and function of circulating monocytes, proinflammatory cytokine levels, gut barrier damage, and bacterial translocation in chronic spinal cord injury (SCI) patients. Thus, 56 SCI patients and 28 healthy donors were studied. The levels of circulating CD14+highCD16−, CD14+highCD16+, and CD14+lowCD16+ monocytes, membrane TLR2, TLR4, and TLR9, phagocytic activity, ROS generation, and intracytoplasmic TNF-α, IL-1, IL-6, and IL-10 after lipopolysaccharide (LPS) stimulation were analyzed by polychromatic flow cytometry. Serum TNF-α, IL-1, IL-6 and IL-10 levels were measured by Luminex and LPS-binding protein (LBP), intestinal fatty acid-binding protein (I-FABP) and zonulin by ELISA. SCI patients had normal monocyte counts and subset distribution. CD14+highCD16− and CD14+highCD16+ monocytes exhibited decreased TLR4, normal TLR2 and increased TLR9 expression. CD14+highCD16− monocytes had increased LPS-induced TNF-α but normal IL-1, IL-6, and IL-10 production. Monocytes exhibited defective phagocytosis but normal ROS production. Patients had enhanced serum TNF-α and IL-6 levels, normal IL-1 and IL-10 levels, and increased circulating LBP, I-FABP, and zonulin levels. Chronic SCI patients displayed impaired circulating monocyte function. These patients exhibited a systemic proinflammatory state characterized by enhanced serum TNF-α and IL-6 levels. These patients also had increased bacterial translocation and gut barrier damage. 相似文献
