CAR (Chimeric Antigen Receptor) T cells have demonstrated clinical success for the treatment of multiple lymphomas and leukaemias, but not for various solid tumours, despite promising data from murine models. Lower effective CAR T-cell delivery rates to human solid tumours compared to haematological malignancies in humans and solid tumours in mice might partially explain these divergent outcomes. We used anatomical and physiological data for human and rodent circulatory systems to calculate the typical perfusion of healthy and tumour tissues, and estimated the upper limits of immune cell delivery rates across different organs, tumour types and species. Estimated maximum delivery rates were up to 10 000-fold greater in mice than humans yet reported CAR T-cell doses are typically only 10–100-fold lower in mice, suggesting that the effective delivery rates of CAR T cells into tumours in clinical trials are far lower than in corresponding mouse models. Estimated delivery rates were found to be consistent with published positron emission tomography data. Results suggest that higher effective human doses may be needed to drive efficacy comparable to mouse solid tumour models, and that lower doses should be tested in mice. We posit that quantitation of species and organ-specific delivery and homing of engineered T cells will be key to unlocking their potential for solid tumours.  相似文献

The most complete form of academic timetabling problem is the population and course timetabling problem. In this problem, there may be multiple classes of each subject, and the decision on which students are to constitute each class is made in concert with the decision on the timetable for each class. In order to solve this problem, it is normally simplified or decomposed in some fashion. One simplification commonly used in practice is known as blocking: it is assumed that the classes can be partitioned into sets of classes (or blocks) that will be timetabled in parallel. This restricts clashing to occur only between classes in the same block, and essentially removes the timetabling aspect of the problem, which can be carried out once the blocks are constituted and the classes populated. The problem of constituting the blocks and populating the classes, known as the course blocking and population problem, is nevertheless a challenging problem, and provides the focus of this paper. We demonstrate, using data provided by a local high school, that integer linear programming approaches can solve the problem in a matter of seconds. Key features include remodelling to remove symmetry caused by students with identical subject selection, and the observation that in practice, only integrality of the block composition variables needs to be enforced; the class population aspects of the model have strong integrality properties.  相似文献

A number of recent initiatives in both academia and industry have sought to achieve improvements in e-businesses through the utilization of Business Process Management (BPM) methodologies and tools. However there are still some inadequacies that need to be addressed when it comes to achieving alignment between business goals and business processes. The User Requirements Notation (URN), recently standardized by ITU-T, has some unique features and capabilities beyond what is available in other notations that can help address alignment issues. In this paper, a URN-based framework and its supporting toolset are introduced which provide business process monitoring and performance management capabilities integrated across the BPM lifecycle. The framework extends the URN notation with Key Performance Indicators (KPIs) and other concepts to measure and align processes and goals. An example process for controlling access to a healthcare data warehouse is used to illustrate and evaluate the framework. Early results indicate the feasibility of the approach.  相似文献

Given recent experimental results suggesting that neural circuits may evolve through multiple firing states, we develop a framework for estimating state-dependent neural response properties from spike train data. We modify the traditional hidden Markov model (HMM) framework to incorporate stimulus-driven, non-Poisson point-process observations. For maximal flexibility, we allow external, time-varying stimuli and the neurons' own spike histories to drive both the spiking behavior in each state and the transitioning behavior between states. We employ an appropriately modified expectation-maximization algorithm to estimate the model parameters. The expectation step is solved by the standard forward-backward algorithm for HMMs. The maximization step reduces to a set of separable concave optimization problems if the model is restricted slightly. We first test our algorithm on simulated data and are able to fully recover the parameters used to generate the data and accurately recapitulate the sequence of hidden states. We then apply our algorithm to a recently published data set in which the observed neuronal ensembles displayed multistate behavior and show that inclusion of spike history information significantly improves the fit of the model. Additionally, we show that a simple reformulation of the state space of the underlying Markov chain allows us to implement a hybrid half-multistate, half-histogram model that may be more appropriate for capturing the complexity of certain data sets than either a simple HMM or a simple peristimulus time histogram model alone.  相似文献

Pattern selection methods have been traditionally developed with a dependency on a specific classifier. In contrast, this paper presents a method that selects critical patterns deemed to carry essential information applicable to train those types of classifiers which require spatial information of the training data set. Critical patterns include those edge patterns that define the boundary and those border patterns that separate classes. The proposed method selects patterns from a new perspective, primarily based on their location in input space. It determines class edge patterns with the assistance of the approximated tangent hyperplane of a class surface. It also identifies border patterns between classes using local probability. The proposed method is evaluated on benchmark problems using popular classifiers, including multilayer perceptrons, radial basis functions, support vector machines, and nearest neighbors. The proposed approach is also compared with four state-of-the-art approaches and it is shown to provide similar but more consistent accuracy from a reduced data set. Experimental results demonstrate that it selects patterns sufficient to represent class boundary and to preserve the decision surface.  相似文献

In the st Bounded Leg Shortest Path (stBLSP) problem we are given two points s and t and a real number L, and are required to compute an L-bounded leg shortest path from s to t. In the All-Pairs Bounded Leg Shortest Path (apBLSP) problem we are required to build a data structure that, given any two query points from V and a real number L, outputs the length of the L-bounded leg shortest path (a distance query) or the path itself (a path query). Recent developments in multi-electrode recordings enable the simultaneous measurement of the spiking activity of many neurons. Analysis of such multineuronal data is one of the key challenge in computational neuroscience today. In this work, we develop a multivariate point-process model in which the observed activity of a network of neurons depends on three terms: (1) the experimentally-controlled stimulus; (2) the spiking history of the observed neurons; and (3) a hidden term that corresponds, for example, to common input from an unobserved population of neurons that is presynaptic to two or more cells in the observed population. We consider two models for the network firing-rates, one of which is computationally and analytically tractable but can lead to unrealistically high firing-rates, while the other with reasonable firing-rates imposes a greater computational burden. We develop an expectation-maximization algorithm for fitting the parameters of both the models. For the analytically tractable model the expectation step is based on a continuous-time implementation of the extended Kalman smoother, and the maximization step involves two concave maximization problems which may be solved in parallel. The other model that we consider necessitates the use of Monte Carlo methods for the expectation as well as maximization step. We discuss the trade-off involved in choosing between the two models and the associated methods. The techniques developed allow us to solve a variety of inference problems in a straightforward, computationally efficient fashion; for example, we may use the model to predict network activity given an arbitrary stimulus, infer a neuron's ring rate given the stimulus and the activity of the other observed neurons, and perform optimal stimulus decoding and prediction. We present several detailed simulation studies which explore the strengths and limitations of our approach.  相似文献

The field of interaction design to date has been predominantly concerned with designing products, that is, devices, systems, and more recently services. A growing body of theoretical and empirical analyses suggests that the scope of interaction design needs to be expanded: An explicit concern of the field should include not only helping designers create better products but also helping people themselves create better environments for their work, learning, and leisure activities. In this article we argue that expanding the scope of interaction design beyond products requires a revision of some of the most central concepts in interaction design, including the notion of “the object of design” and our understanding of the impact of technologies on human practices. The aim of the article is to explore some of these conceptual challenges and discuss possible ways of dealing with them. We differentiate between intrinsic and extrinsic technology-enabled practice transformation, and foreground the need for interaction design research and practice to more directly deal with analysis and construction of technology-enhanced activity spaces. The implications of these notions for the research agenda of interaction design are discussed.  相似文献
