The regulation of cell volume is an essential function that is coupled to a variety of physiological processes such as receptor recycling, excitability and contraction, cell proliferation, migration, and programmed cell death. Under stress, cells undergo emergency swelling and respond to such a phenomenon with a regulatory volume decrease (RVD) where they release cellular ions, and other osmolytes as well as a concomitant loss of water. The link between P-glycoprotein, a transmembrane transporter, and cell volume regulation is controversial, and changes in cells volume are measured using microscopy or electrophysiology. For instance, by using the patch-clamp method, our team demonstrated that chloride currents activated in the RVD were more intense and rapid in a breast cancer cell line overexpressing the P-glycoprotein (P-gp). The Cell Lab Quanta SC is a flow cytometry system that simultaneously measures electronic volume, side scatter and three fluorescent colors; altogether this provides unsurpassed population resolution and accurate cell counting. Therefore, here we propose a novel method to follow cellular volume. By using the Coulter-type channel of the cytometer Cell Lab Quanta SC MPL (multi-platform loading), we demonstrated a role for the P-gp during different osmotic treatments, but also a differential activity of the P-gp through the cell cycle. Altogether, our data strongly suggests a role of P-gp in cell volume regulation.  相似文献

The state of Nevada has extensive mineral resources, and is the largest producer of gold in the USA as well as fourth in world gold production. Mercury (Hg) is often present in the hydrothermal systems that produce gold deposits, and can be found in elevated concentrations in gold ore. As a result, mining of gold ore in Nevada has been shown to release Hg to the atmosphere from point and non-point sources. This project focused on measurement of air-soil Hg exchange associated with undisturbed soils and bedrock outcrops in the vicinity of two large gold mines. Field and laboratory data collected were used to identify the important variables controlling Hg flux from these surfaces, and to estimate a net flux from the areas adjacent to the active mines as well as that occurring from the mined area pre-disturbance. Mean daily flux by substrate type ranged from 9 ng m− 2 day− 1 to 140 ng m− 2 day− 1. Periods of net deposition of elemental Hg were observed when air masses originating from a mine site moved over sampling locations. Based on these observations and measured soil Hg concentrations we suggest that emissions from point and non-point sources at the mines are a source of Hg to the surrounding substrates with the amount deposited not being of an environmental concern but of interest mainly with respect to the cycling of atmospheric elemental Hg. Observations indicate that while some component of the deposited Hg is sequestered in the soil, this Hg is gradually released back to the atmosphere over time. Estimated pre-disturbance emissions from the current mine footprints based on field data were 0.1 and 1.7 kg yr− 1, compared to that estimated for the current non-point mining sources of 19 and 109 kg yr− 1, respectively.  相似文献

The moisture balance at room scale is influenced by the air change rate, moisture production and vapour transfer with the envelope. However, results may differ strongly from one study to another depending on the sizing of the ventilation system, the boundary conditions and the modelling of vapour transfer in the walls. This paper aims to provide a realistic comparison of usual construction techniques based on energy consumption, indoor comfort and durability. To achieve this objective, an existing whole-building heat–air–moisture simulation tool was selected to compute coupled transfer at room scale over an entire year. Moisture production due to occupancy was modelled using a stochastic approach. Four cases were selected to emphasize the differences of both a vapour-permeable wall assembly and a relative humidity sensitive ventilation system compared to common practices.  相似文献

Earth to air heat exchanger (EAHE) is a well‐known technique used to preheat or precool outdoor air before blowing it into a building. However, its geometry is often very simple as it consists in one or multiple straight pipes, while more complex arrangements can be found in heat exchangers design. In this paper, we explore the advantage of designing an EAHE as a network through the Constructal law point of view. A methodology is first proposed to design a single pipe EAHE when the need is defined in terms of cooling power, overall efficiency and enthalpy difference between the inlet air and the ground. Next, the single pipe EAHE is used as a reference for designing a tree‐shaped network under the constraint of identical fluid volume and cooling power. The geometrical features are allowed to change for the different branches of the network. The network coefficient of performance is found to increase significantly with the bifurcation level, illustrating the superior performances of the network. This approach was found to be robust as the improvements were not depending on the cooling demand or the environmental conditions. However, further work is needed to move from this theoretical result to practical considerations.  相似文献

The management of end-of-life systems is becoming a major concern for systems manufacturers, as the negative impact of these systems on the environment is a matter of increasing public awareness, and their appropriate treatment offers economic opportunities. In this context, the disassembly of these systems in order to recycle their components is a possible and sound option that can make it possible to sustain economical progress while respecting environment requirements. The work undertaken in this paper considers modelling and optimising issues of such disassembly activities. An integrated approach is proposed to model and optimise the selection of valuable components of end-of-life systems, their recycling options and the way to obtain them. Because the framework of such problems is highly uncertain, we propose the use of Bayesian networks and their extension in terms of influence diagrams as mathematical tools for structuring and managing uncertainties. With this approach, one can take into account uncertainties rising from different sources on one hand and as a support for optimisation on the other hand.  相似文献

Microscopic sessile suspension feeders live attached to surfaces and, by consuming bacteria-sized prey and by being consumed, they form an important part of aquatic ecosystems. Their environmental impact is mediated by their feeding rate, which depends on a self-generated feeding current. The feeding rate has been hypothesized to be limited by recirculating eddies that cause the organisms to feed from water that is depleted of food particles. However, those results considered organisms in still water, while ambient flow is often present in their natural habitats. We show, using a point-force model, that even very slow ambient flow, with speed several orders of magnitude less than that of the self-generated feeding current, is sufficient to disrupt the eddies around perpendicular suspension feeders, providing a constant supply of food-rich water. However, the feeding rate decreases in external flow at a range of non-perpendicular orientations due to the formation of recirculation structures not seen in still water. We quantify the feeding flow and observe such recirculation experimentally for the suspension feeder Vorticella convallaria in external flows typical of streams and rivers.  相似文献

This paper presents an adaptation and extension of the shock filters on weighted graphs using the formalism of partial difference equations. This adaptation leads to new morphological operators that alternate between nonlocal dilation and nonlocal erosion filter type on graphs. Furthermore, this adaptation extends the shock filter applications to any data that can be represented by graphs. This approach is illustrated through image, data defined on region maps, and 3D point cloud processing.  相似文献
