Tumor cell aggregation is critical for cell survival following the loss of extracellular matrix attachment and dissemination. However, the underlying mechanotransduction of clustering solitary tumor cells is poorly understood, especially in non-small cell lung cancers (NSCLC). Here, we examined whether cell surface protrusions played an important role in facilitating the physical contact between floating cells detached from a substrate. We employed poly-2-hydroxyethyl methacrylate-based 3D culture methods to mimic in vivo tumor cell cluster formation. The suprastructural analysis of human NSCLC A549 cell spheroids showed that finger-like protrusions clung together via the actin cytoskeleton. Time-lapse holotomography demonstrated that the finger-like protrusions of free-floating cells in 3D culture displayed exploratory coalescence. Global gene expression analysis demonstrated that the genes in the organic hydroxyl transport were particularly enriched in the A549 cell spheroids. Particularly, the knockdown of the water channel aquaporin 3 gene (AQP3) impaired multicellular aggregate formation in 3D culture through the rearrangement of the actomyosin cytoskeleton. Moreover, the cells with reduced levels of AQP3 decreased their transmigration. Overall, these data indicate that cell detachment-upregulated AQP3 contributes to cell surface protrusions through actomyosin cytoskeleton remodeling, causing the aggressive aggregation of free-floating cells dependent on the property of the substratum and collective metastasis.  相似文献

Automatic spacing in Korean is used to correct spacing units in a given input sentence. The demand for automatic spacing has been increasing owing to frequent incorrect spacing in recent media, such as the Internet and mobile networks. Therefore, herein, we propose a transformer encoder that reads a sentence bidirectionally and can be pretrained using an out-of-task corpus. Notably, our model exhibited the highest character accuracy (98.42%) among the existing automatic spacing models for Korean. We experimentally validated the effectiveness of bidirectional encoding and pretraining for automatic spacing in Korean. Moreover, we conclude that pretraining is more important than fine-tuning and data size.  相似文献

The performance of lead-halide perovskite light-emitting diodes (LEDs) has increased rapidly in recent years. However, most reports feature devices operated at relatively small current densities (<500 mA cm−2) with moderate radiance (<400 W sr−1 m−2). Here, Joule heating and inefficient thermal dissipation are shown to be major obstacles toward high radiance and long lifetime. Several thermal management strategies are proposed in this work, such as doping charge-transport layers, optimizing device geometry, and attaching heat spreaders and sinks. Combining these strategies, high-performance perovskite LEDs are demonstrated with maximum radiance of 2555 W sr−1 m−2, peak external quantum efficiency (EQE) of 17%, considerably reduced EQE roll-off (EQE > 10% to current densities as high as 2000 mA cm−2), and tenfold increase in operational lifetime (when driven at 100 mA cm−2). Furthermore, with proper thermal management, a maximum current density of 2.5 kA cm−2 and an EQE of ≈1% at 1 kA cm−2 are shown using electrical pulses, which represents an important milestone toward electrically driven perovskite lasers.  相似文献

The surface energies of pristine multi-walled carbon nanotubes (MWCNTs) and MWCNTs functionalized with carboxylic acid (MWCNT-COOH), acyl chloride and ethyl amine were characterized, and the effects of the changes in MWCNT surface energies on the interfacial adhesion and reinforcement of the composites were explored. When the surface energy of pristine MWCNTs was compared to that of functionalized MWCNTs, a decrease in the dispersive surface energy and an increase in the polar surface energy were observed. Interfacial adhesion energies between MWCNTs and various polymers were estimated from surface energy values of MWCNTs and various polymers. Among the MWCNTs, polyethylene, polystyrene and bisphenol-A polycarbonate (PC) had the highest interfacial energy with pristine MWCNTs, while nylon 6,6 and polyacrylamine exhibited the highest interfacial energy with MWCNT-COOH. When tensile properties and adhesion at the interface of PC and nylon 6,6 composites containing MWCNTs were examined, composites having high interfacial adhesion energy exhibited greater adhesion at the interface and reinforcement.  相似文献

Sphingolipids are crucial for the life of the cell. In land‐dwelling mammals, they are equally important outside the cell—in the extracellular space of the skin barrier—because they prevent loss of water. Although a large body of research has elucidated many of the functions of sphingolipids, their extensive structural diversity remains intriguing. A new class of sphingolipids based on 6‐hydroxylated sphingosine has recently been identified in human skin. Abnormal levels of these 6‐hydroxylated ceramides have repeatedly been observed in atopic dermatitis; however, neither the biosynthesis nor the roles of these unique ceramide subclasses have been established in the human body. In this Minireview, we summarize the current knowledge of 6‐hydroxyceramides, including their discovery, structure, stereochemistry, occurrence in healthy and diseased human epidermis, and synthetic approaches to 6‐hydroxysphingosine and related ceramides.  相似文献

Low temperature water–gas shift (WGS) reaction has been carried out at the gas hourly space velocity of 72,152 h−1 over Cu–CeO2 catalyst prepared by a co-precipitation method. Cu loading was optimized to obtain highly active co-precipitated Cu–CeO2 catalysts for low temperature WGS. 80 wt% Cu–CeO2 exhibited the highest CO conversion as well as the most stable activity (XCO > 46% at 240 °C for 100 h). The excellent catalytic performance is mainly due to a strong metal to support interaction, resulting in the prevention of Cu sintering.  相似文献

In temporal data analysis, noisy data is inevitable in both testing and training. This noise can seriously influence the performance of the temporal data analysis. To address this problem, we propose a novel method, termed Selective Temporal Filtering that builds a noise-free model for classification during training and identifies key-feature vectors that are noise-filtered data from the input sequence during testing. The use of these key-feature vectors makes the classifier robust to noise within the input space. The proposed method is validated on a synthetic-dataset and a database of American Sign Language. Using key-feature vectors results in robust performance with respect to the noise content. Futhermore, we are able to show that the proposed method not only outperforms Conditional Random Fields and Hidden Markov Models in noisy environments, but also in a well-controlled environment where we assume no significant noise vectors exist.  相似文献
