Cancer is going to be the first cause of mortality worldwide in the 21th century. It is considered a multifactorial disease that results from the combined influence of many genetic aberrations, leading to abnormal cell proliferation. As microtubules are strongly implicated in cellular growth, they represent an important target for cancer treatment. The well-known microtubule-targeting agents (MTAs) including paclitaxel, colchicine and vinca alkaloids are commonly used in the treatment of various cancers. However, adverse effects and drug resistance are major limitations in their clinical use. To find new candidates able to induce microtubule alteration with reduced toxic effects or drug resistance, we studied a small new series of derivatives that present imidazolinic, guanidinic, thioureidic and hydrazinic groups ( 1 – 9 ). All the compounds were tested for their antitumor activity against a panel of six tumoral cell models. In particular, compound 8 (nonane-1,9-diyl-bis-S-amidinothiourea dihydrobromide) showed the lowest IC50 value against HeLa cells, together with a low cytotoxicity for normal cells. This compound was able to induce the apoptotic mitochondrial pathway and inhibited tubulin polymerization with a similar efficacy to vinblastine and nocodazole. Taken together, these promising biological properties make compound 8 useful for the development of novel therapeutic approaches in cancer treatment. 相似文献

Resveratrol (RSV) is a natural compound that displays several pharmacological properties, including anti-cancer actions. However, its clinical application is limited because of its low solubility and bioavailability. Here, the antiproliferative and anti-inflammatory activity of a series of phenylacetamide RSV derivatives has been evaluated in several cancer cell lines. These derivatives contain a monosubstituted aromatic ring that could mimic the RSV phenolic nucleus and a longer flexible chain that could confer a better stability and bioavailability than RSV. Using MTT assay, we demonstrated that most derivatives exerted antiproliferative effects in almost all of the cancer cell lines tested. Among them, derivative 2, that showed greater bioavailability than RSV, was the most active, particularly against estrogen receptor positive (ER+) MCF7 and estrogen receptor negative (ER-) MDA-MB231 breast cancer cell lines. Moreover, we demonstrated that these derivatives, particularly derivative 2, were able to inhibit NO and ROS synthesis and PGE2 secretion in lipopolysaccharide (LPS)-activated U937 human monocytic cells (derived from a histiocytoma). In order to define the molecular mechanisms underlying the antiproliferative effects of derivative 2, we found that it determined cell cycle arrest at the G1 phase, modified the expression of cell cycle regulatory proteins, and ultimately triggered apoptotic cell death in both breast cancer cell lines. Taken together, these results highlight the studied RSV derivatives, particularly derivative 2, as promising tools for the development of new and more bioavailable derivatives useful in the treatment of breast cancer. 相似文献

To train Neural Networks (NNs) in a supervised way, estimations of an objective function must be carried out. The value of
this function decreases as the training progresses and so, the number of test observations necessary for an accurate estimation
has to be increased. Consequently, the training computational cost is unaffordable for very low objective function value estimations,
and the use of Importance Sampling (IS) techniques becomes convenient. The study of three different objective functions is
considered, which implies the proposal of estimators of the objective function using IS techniques: the Mean-Square error,
the Cross Entropy error and the Misclassification error criteria. The values of these functions are estimated by IS techniques,
and the results are used to train NNs by the application of Genetic Algorithms. Results for a binary detection in Gaussian
noise are provided. These results show the evolution of the parameters during the training and the performances of the proposed
detectors in terms of error probability and Receiver Operating Characteristics curves. At the end of the study, the obtained
results justify the convenience of using IS in the training. 相似文献

This paper describes the preparation of a hydrophilic copolymer membrane with two comonomers: 1-vinyl-2-pyrrolidone (commercial) and a new water insoluble piperazinedione derivative, which is a fluorogenic mercury sensing motif. The dense membrane permitted the fluorogenic detection of Hg2+ in aqueous media. The sensitivity of the membrane this cation was significantly high, with a detection limit of 10 ppt. This limit of detection is much lower than that of DMSO/water solutions of the sensing monomer, which is 10 ppb, and significantly lower than the maximum contaminant level (EPA) in drinking water, 2 ppb. 相似文献

OBJECTIVE: The measurements of transient evoked otoacoustic emissions and distortion-product otoacoustic emissions are being used increasingly, both as an objective hearing test clinically, and as a research tool to investigate the micromechanical aspects of cochlear function. We hypothesized that localized damage in the apical or middle cochlear turns may have an influence on the micromechanics and the function of adjacent, apparently normal cochlea. For that purpose, we used an animal model of localized apical and middle-turn cochlear lesions. METHOD: Extent of damage was assessed by scanning electron microscopy and the function of the damaged cochlea by change in the otoacoustic emission (OAE) levels. RESULTS: We found that localized damage to the apical or middle turn may be accompanied by an increase in OAE measured from adjacent apparently normal cochlea. CONCLUSION: Explanations to this phenomenon are suggested, and possible clinical associations such as to Meniere's disease and to sudden hearing loss are reviewed. 相似文献

Meeting, to discuss and share information, take decisions and allocate tasks, is a central aspect of human activity. Computer mediated communication offers enhanced possibilities for synchronous collaboration by allowing seamless capture of meetings, thus relieving participants from time-consuming documentation tasks. However, in order for meeting systems to be truly effective, they must allow users to efficiently navigate and retrieve information of interest from recorded meetings. In this article, we review the state of the art in multimedia segmentation, indexing and browsing techniques and show how existing meeting browser systems build on these techniques and integrate various modalities to meet their users’ information needs. 相似文献

This paper provides an analysis of the collaborative work conducted at a multidisciplinary medical team meeting, where a patient’s
definitive diagnosis is agreed, by consensus. The features that distinguish this process of diagnostic work by consensus are
examined in depth. The current use of technology to support this collaborative activity is described, and experienced deficiencies
are identified. Emphasis is placed on the visual and perceptual difficulty for individual specialities in making interpretations,
and on how, through collaboration in discussion, definitive diagnosis is actually achieved. The challenge for providing adequate
support for the multidisciplinary team at their meeting is outlined, given the multifaceted nature of the setting, i.e. patient
management, educational, organizational and social functions, that need to be satisfied.

The design, synthesis, crystal structure and interaction with DNA of the N,N'-(butane-1,4-diyl)bis(guanidinium) tetrachloroplatinate(ll) are described. Crystal data: a = 8.152(1), b = 8.889(4), c = 10.700(3) A , alpha = 81.59(3), beta = 87.99(5), gamma = 78.48(6) degrees , V = 752(1) A(3), Z = 2 , space group P-1. The structure was refined to R = 0.039 and Rw = 0.046 from 1853 reflections (I > 3sigma(I)). This compound, named PtC(4)Gua, does not exhibit a center of symmetry and the center linker chain C(2) - C(3) - C(4) - C(5) is in gauche conformation. The cation is bisprotonated with the H(+) attached to the imine group of each terminal guanidinium function. The presence of the platinum moiety reinforces the binding of the butane(bis)guanidinium structure with double stranded DNA as judged from thermal denaturation studies and DNA unwinding experiments. 相似文献
