Femtosecond pulses from a Ti:Sapphire laser were used to irradiate specimens of yttria-stabilised (35% mol) tetragonal zirconia (Y-TZP) with the purpose of studying the effects of the irradiations on their surface properties and morphology after ageing. Zirconia disks were divided into eight groups (n = 32) according to their surface treatment and subsequent ageing: Control: no treatment; sandblasting: Al2O3 sandblasting 50 μm; and ultrashort laser pulses irradiation with 25 μJ pulses, considering two different scanning steps based on the width between two grooves. These groups were duplicated and submitted to ageing. The surfaces were analysed using scanning electron microscopy (SEM), and X-ray diffraction. A finite element analysis, a biaxial flexure test, as well as fractographic and Weibull analyses, were performed. The strengths of the disks were statistically different for the treatment factor, and the principal stresses seemed to be concentrated at the centre of the specimens, as predicted by the computer simulations. Ageing decreased the strengths for all groups and increased the Weibull modulus for the laser group with the 40 μm-width between two grooves. The sandblasting group presented the highest monoclinic phase peak. Although the most significant strength was found within the sandblasting group, the phase transformation was favourable to the laser groups. The Weibull modulus was higher for the laser group with the 60 μm-width between two grooves, confirming the highest homogeneity of its failure distribution. Regardless of the surface treatment, strength was decreased with ageing in all groups. The femtosecond Ti:Sa ultra-short pulse laser irradiation can be suggested as an alternative to the gold standard sandblasting in long-term Y-TZP zirconia rehabilitations, such as crowns and veneers. 相似文献
Protein trafficking is altered when normal cells acquire a tumor phenotype. A key subcellular compartment in regulating protein trafficking is the Golgi apparatus, but its role in carcinogenesis is still not well defined. Golgi phosphoprotein 3 (GOLPH3), a peripheral membrane protein mostly localized at the trans-Golgi network, is overexpressed in several tumor types including glioblastoma multiforme (GBM), the most lethal primary brain tumor. Moreover, GOLPH3 is currently considered an oncoprotein, however its precise function in GBM is not fully understood. Here, we analyzed in T98G cells of GBM, which express high levels of epidermal growth factor receptor (EGFR), the effect of stable RNAi-mediated knockdown of GOLPH3. We found that silencing GOLPH3 caused a significant reduction in the proliferation of T98G cells and an unexpected increase in total EGFR levels, even at the cell surface, which was however less prone to ligand-induced autophosphorylation. Furthermore, silencing GOLPH3 decreased EGFR sialylation and fucosylation, which correlated with delayed ligand-induced EGFR downregulation and its accumulation at endo-lysosomal compartments. Finally, we found that EGF failed at promoting EGFR ubiquitylation when the levels of GOLPH3 were reduced. Altogether, our results show that GOLPH3 in T98G cells regulates the endocytic trafficking and activation of EGFR likely by affecting its extent of glycosylation and ubiquitylation. 相似文献
The synthesis of thiazolyl‐7‐aminocephalosporanic acid, a pharmaceutical precursor of the β‐lactam antibiotic drug cefodizime, could be successfully realized in a continuous microstructured flow system. This was accomplished by changing the process window from batch, having a reaction temperature of 60 °C with a reaction time of 1 h, to a flow system at temperatures > 100 °C and reaction times of several minutes. In the dipteran genus Hermetia, only 6 of the 78 valid species have documented immature stages: H. albitarsis Fabricius, 1805, H. aurata Bellardi, 1859, H. concinna Williston, 1900, H. illucens (Linnaeus, 1758), H. panamensis Greene, 1940 and H. pulchra Weidemann, 1830. In particular, H. illucens stands out due to its reported applicability for forensic, medical and economic purposes. Here, we described the morphology of eggs and immature stages of this species, with a view to detecting differences between instars and in the pupal stage, which should eventually help properly identifying larval age. We utilized both optical and scanning electron microscopy tools. The eggs are elliptical and elongated, and color varies from cream white to yellowish. The larvae are apodal, hemichephalic and holopneustic, flattened dorso‐ventrally and may be recognized by the head elongated, dorsal and ventral chaetotaxy of the cephalic capsule, thoracic and abdominal segments, and the morphology of the anterior and posterior spiracles. The pupae are adecticous and coarctate, tegument dark brown and pruinescence varying from brown to golden. The overall morphology across instars is similar, but marked variations were observed in the shape of the antennal articuli and the shape of the setae (first instar compared to the others). Our results supplement the biological information on Hermetia illucens and should aid the proper identification and aging of juveniles in the field, as a way to minimize errors in the calculation of the post‐mortem interval. 相似文献
In the context of the high-level radioactive waste disposal CIGEO, the corrosion rate due to microbially influenced corrosion (MIC) has to be evaluated. In France, it is envisaged to dispose of high- and intermediate-level long-lived radioactive waste at a depth of 500 m in a deep geological disposal, drilled in the Callovo-Oxfordian claystone (Cox) formation. To do so, a carbon steel casing will be inserted inside disposal cells, which are horizontal tunnels drilled in the Cox. A specific cement grout will be injected between the carbon steel casing and the claystone. A study was conducted to evaluate the possibility of MIC on carbon steel in the foreseeable high radioactive waste disposal. The corrosiveness of various environments was investigated at 50°C and 80°C with or without microorganisms enriched from samples of Andra's underground research laboratory. The monitoring of corrosion during the experiments was ensured using gravimetric method and real-time corrosion monitoring using sensors based on the measurements of the electrical resistance. The corrosion data were completed with microbiological analyses including cultural and molecular characterizations. 相似文献
This research aims to illustrate the potential use of concepts, techniques, and mining process tools to improve the systematic review process. Thus, a review was performed on two online databases (Scopus and ISI Web of Science) from 2012 to 2019. A total of 9649 studies were identified, which were analyzed using probabilistic topic modeling procedures within a machine learning approach. The Latent Dirichlet Allocation method, chosen for modeling, required the following stages: 1) data cleansing, and 2) data modeling into topics for coherence and perplexity analysis. All research was conducted according to the standards of the Preferred Reporting Items for Systematic Reviews and Meta-Analyses in a fully computerized way. The computational literature review is an integral part of a broader literature review process. The results presented met three criteria: (1) literature review for a research area, (2) analysis and classification of journals, and (3) analysis and classification of academic and individual research teams. The contribution of the article is to demonstrate how the publication network is formed in this particular field of research, and how the content of abstracts can be automatically analyzed to provide a set of research topics for quick understanding and application in future projects.
