Amphotericin B (AMB) was often used in intra-articular injection administration for fungal arthritis, because it could often bring a satisfactory therapeutic efficacy and a minimum systemic toxic side effect. However, because of the multiple operations and the frequent injections, the compliance of the patients was bad. Therefore, to develop a long-term sustained-released preparation of AMB for mycotic arthritis intra-articular administration is of great significance. The purpose of present study was to develop a long-term sustained-released in situ gel of a water-insoluble drug AMB for mycotic arthritis intra-articular administration. Based on the evaluations of the in vitro properties of the formulations, the formulation containing 10% (w/w) ethanol, 15% (w/w) PG, 0.75% (w/w) HA, 5% (w/w) purified soybean oil, 0.03% (w/w) α-tocopherol, 15% (w/w) water and 55% (w/w) glyceryl monooleate was selected as a suitable intra-articular injectable in situ gel drug delivery system for water-insoluble drug AMB. Furthermore, the results of the in vivo study on rabbits showed that the selected formulation was a safe and effective long-term sustained-released intra-articular injectable AMB preparation. Therefore, the presented in situ AMB gel could reduce the frequency of the administration in the AMB treatment of fungal arthritis, and then would get a good patient compliance.  相似文献

Structural isomers of monoacylglycerols (monoglycerides, MAGs) were identified and compared after degradation of butter oil by two strains of Penicillium roquefortii and a commercial lipase from P roquefortii (EC 3.1.1.3) at pH 7.0 and 10 °C. The conditions were selected as they were comparable with those used in the manufacture of blue mould‐ripened cheese. The commercial lipase was selected to compare with the fungal strains in terms of acyl migration. Results showed that the main isomers formed by lipolysis with the commercial lipase were sn‐2 MAGs (64 mol%), whilst spores and emerging mycelia of P roquefortii produced mainly sn‐1(3) MAGs (83–90 mol%). The work reported here may lead to further assessment of different MAG structural isomers as natural preservatives in foods and dairy products. © 2002 Society of Chemical Industry  相似文献

Production of the fungus Aspergillus niger NRRL 330 was studied in submerged fermentation with ram horn hydrolysate (RHH) as substrate. The characteristics of RHH have been reported previously. The RHH was enriched by addition of glucose and KH2PO4. The effects of kinetic parameters on the biomass yield of the fungus were investigated. The optimal conditions for growth of A niger on RHH were initial pH 6.5, temperature 30 °C, fermentation time 96 h and agitation speed 150 rpm. Under these optimal conditions the initial carbohydrate content of RHH was reduced from 1.52 to 0.2% and the biomass yield was 8.9 g l?1. The biomass contained about 48.1% protein, 5.2% fat and 9.4% ash (on a dry weight basis). The amino acid content of the biomass was compared with Food and Agricultural Organisation (FAO) and animal feed standards. The protein produced contained all the essential amino acids for animal feed, but the amounts of these amino acids were somewhat lower than those of FAO and soybean reference protein. However, the amino acid composition of the biomass was better than that of animal feed. The results with RHH were also compared with previously reported data on fungal mycelium grown on waste liquor substrate. The sorption of toxic metals by fungal mycelia grown in clay‐containing medium is reported in this work. Biomass of melanin‐producing microfungi of the genus Cladosporium (C cladosporioides, C resinae and C herbarum) and Aureobasidium pullulans, clay minerals and fungal mycelia grown in clay‐containing medium were compared for their equilibrium Cu and Cd uptake from pH‐buffered solutions using experimental sorption isotherms. Bentonite (Cherkassy) in the natural form was shown to be the best Cu sorbent compared with the other clays and clay minerals tested. Cu sorption isotherms fitted the Langmuir sorption model for all cases. Comparison of Cu and Cd uptake for biomass and bentonite showed a lower biomass uptake capacity but a considerably higher affinity of the biosorbent for the solute. The presence of kaolinite and palygorskite in the medium generally reduced both Cu and Cd sorption capacity and the metal‐binding ability of the fungal–clay mixtures. In contrast, addition of bentonite into the medium did not appreciably alter the Cd sorption ability but increased the sorption of Cu by A pullulans and C cladosporioides grown in this medium. A common feature for all fungi grown in the presence of bentonite was an increase in the Cu sorption capacity (Qmax) of the biomineral sorbents and a reduction in their affinity (Langmuir parameter b) compared with control biomass. A difference between predicted and experimental data obtained for biomass grown on bentonite medium was also observed. The connection between the sorption capacity of biomass grown in clay‐containing medium, mycelial morphology and the structure of fungal pellets is discussed and a mechanism for the changed sorption capacity of the combined biomineral sorbents is proposed which involves blocking or modification of binding sites on biotic and abiotic components of the ‘biomineral’ association. © 2002 Society of Chemical Industry  相似文献

The origin of olfactory stimuli involved in the host microhabitat location inCotesia flavipes, a parasitoid of stem-borer larvae, was investigated in a Y-tube olfactometer. The response of femaleC. flavipes towards different components of the plant-host complex, consisting of a maize plant infested with two or more larvae of the stem borerChilo partellus, was tested in dualchoice tests. The concealed lifestyle of the stem-borer larvae did not limit the emission of volatiles attractive to a parasitoid. A major source of the attractive volatiles from the plant-host complex was the stem-borer-injured stem, including the frass produced by the feeding larvae. Moreover, the production of volatiles attractive to a parasitoid was not restricted to the infested stem part but occurs systemically throughout the plant. The uninfested leaves of a stem-borer-infested plant were found to emit volatiles that attract femaleC. flavipes. We further demonstrate that an exogenous elicitor of this systemic plant response is situated in the regurgitate of a stem-borer larva. When a minor amount of regurgitate is inoculated into the stem of an uninfested plant, the leaves of the treated plant emit volatiles that attract femaleC. flavipes.  相似文献

The antioxidant vitamin E, -tocopherol, was tested as a candidate elicitor of alterable antiherbivory in soybean plants against cabbage looper larvae. Although a nonspecific antioxidant, vitamin E proved elicitory to the involved sulfhydryl-dependent receptor-energy transducer protein in soybean plasma membrane. Effects of -tocopherol were dependent on dosage, time, and space in the plant. The observed elicited effects were all decreases in herbivory. The best negative phytochemical correlate of looper feeding was the percentage of increased total HPLC peak area of extractables from elicited as compared to nonelicited leaves. Some specific compounds, e.g., glyceollins, were quantitatively major components of the total profile of secondary metabolites.  相似文献

The extraction of oil from fungi (Mortierella ramanniana var.angulispora) was studied using carbon dioxide (CO2), nitrous oxide (N2O), trifluoromethane (CHF3) and sulfur hexafluoride (SF6) under supercritical conditions. The oil solubility was highest in SC-N2O followed by SC-CO2, while both SC-CHF3 and SC-SF6 showed poorer solvent power. The recorded oil solubilities at 333 K and 24.5 MPa were 2.3 wt% in N2O, 0.48 wt% in CO2, 0.0099 wt% in CHF3 and 0.0012 wt% in SF6. The oil solubilities in SC-N2O and SC-CO2 were measured over the pressure range 15.7–29.4 MPa and at temperatures ranging from 313–353 K. N2O always showed greater solvent power than did CO2 at the same temperature and pressure. The solvent power of a supercritical fluid increases with density at a given temperature, and increases with temperature at constant density. The change in neutral lipid composition of the extracted oil with the extraction ratio was measured. Free fatty acids or diglycerides were extracted more easily than triglycerides or sterol esters. The change in fatty acid composition was also measured. The proportion of γ-linolenic acid in the extract remained constant throughout the extraction.  相似文献

The fungal oil extracted fromMortierella ramanniana var.angulispora (IFO 8187) was solvent winterized in order to raise the content of γ-linolenic acid (GLA). Effects of winterization conditions (solvent, oil concentration in the solvent and temperature) and changes of glyceride compositions were discussed. The fungal oil was separated into four diglycerides and 17 triglycerides (TG) with high performance liquid chromatography. The predominant species were POO, POP and LOP, whose contents were 24.4, 22.9 and 9.4% of the total TG, respectively. Ethanol at 4°C gave the highest GLA content of 10.5% in spite of lower yield than with acetone at −20°C. The highest separation efficiency for GLA (ηGLA) was 0.27 with acetone at −20°C and 10% oil concentration, resulting in 8.3% of GLA from the fungal oil at 5.7% LGA. In case of lower oil concentration at 5–20%, ηGLA showed higher in the following order: acetone (−20°C)>n-hexane (−20°C)>acetone (4°C)>petroleum ether (−20°C). The winterization process also proved to be effective for the separation of TG type, Sa2U (Sa; saturated fatty acid; U, unsaturated fatty acid) into the crystallized fraction and SaU2 into the liquid fraction. Acetone at −20°C showed higher separation efficiency for triunsaturated TG than the other solvents.  相似文献

通过向人参毛状根培养基中添加植物激素类物质2,4-二氯苯氧乙酸(2,4-D)、萘乙酸(NAA)、3-吲哚丁酸(IBA)、诱导子茉莉酸甲酯(MJA)和水杨酸(SA),研究了这5种外源物质对人参毛状根的生长量、总皂苷及单体皂苷Rb1的影响。结果表明,3种外源激素在适宜浓度下均能在不同程度上促进总皂苷和单体皂苷Rb1的积累。其中0.5mg/L的IBA能显著促进人参毛状根的生长(月增长量为50.73g/L)和总皂苷(质量分数为3.31%)的积累,单体皂苷Rb1(质量分数为2.293%)的含量也明显增大,使其在总皂苷中所占的比例增大。另外,2种诱导子对人参毛状根的生长无明显促进作用,在适当的浓度下能够促进人参毛状根人参皂苷的积累,并且这两种诱导子对人参皂苷含量的增加具有明显的协同作用,当MJA与SA浓度均为0.001mmol/L时,其对总皂苷和单体皂苷的积累比二者单独作用要大。  相似文献

Fungal endocarditis (FE) is commonly regarded as a rare but fatal disease. The incidence of infective endocarditis (IE) in hemodialysis (HD) patients is thought to be obviously higher than that in the general population. Moreover, IE occurs more likely in HD patients with catheters. With the increase of HD population and extensive use of catheters in HD patients, FE, as a special form of IE, may increase and bring new challenges to clinicians. We reported a case of FE associated with catheter infection in a 44‐year‐old woman on HD. The risk factors and treatment strategies of FE in HD patients were discussed.  相似文献
