Two contrasting investigative techniques provided evidence consistent with the interpretation that female quail (Coturnix japonica) regulate male copulatory behavior by the duration of their immobility and through this behavioral mechanism they also control the rate of fertilization of their eggs. In Experiment 1, copulation tests with males and females from different genetic lines showed that the type of female that participated in a copulatory test significantly influenced the latency of the male's grab, mount, and cloacal contact responses and also determined the efficiency of the male's copulatory behavior. These measures of male performance were correlated with female immobility in Experiment 2, which used a more homogeneous population of quail. Furthermore, 2 of these measures (copulatory efficiency and the latency to make cloacal contact) were correlated with fertilization rate. Xenoantigens cause hyperacute rejection and limit the success of interspecific xenografts. Therefore, genes involved in xenoantigen biosynthesis, such as GGTA1, CMAH, and B4GALNT2, are key targets to improve the outcomes of xenotransplantation. In this study, we introduced a CRISPR/Cas9 system simultaneously targeting GGTA1, CMAH, and B4GALNT2 into in vitro-fertilized zygotes using electroporation for the one-step generation of multiple gene-edited pigs without xenoantigens. First, we optimized the combination of guide RNAs (gRNAs) targeting GGTA1 and CMAH with respect to gene editing efficiency in zygotes, and transferred electroporated embryos with the optimized gRNAs and Cas9 into recipient gilts. Next, we optimized the Cas9 protein concentration with respect to the gene editing efficiency when GGTA1, CMAH, and B4GALNT2 were targeted simultaneously, and generated gene-edited pigs using the optimized conditions. We achieved the one-step generation of GGTA1/CMAH double-edited pigs and GGTA1/CMAH/B4GALNT2 triple-edited pigs. Immunohistological analyses demonstrated the downregulation of xenoantigens; however, these multiple gene-edited pigs were genetic mosaics that failed to knock out some xenoantigens. Although mosaicism should be resolved, the electroporation technique could become a primary method for the one-step generation of multiple gene modifications in pigs aimed at improving pig-to-human xenotransplantation. 相似文献

In the previous paper, we reported the stand growth of slash pine (Pinus elliottii) during the first 11.5 years of plantation in response to (1) initial fertilization at plantation establishment with P rates of 11, 22, 45 and 90 kg P ha–1 which were either banded or broadcast in the presence or absence of basal fertilizers containing 50 kg N ha–1, 50 kg K ha–1 and 5 kg Cu ha–1 and (2) application of additional 40 kg P ha–1 at age 10 years. Here we present the responses in foliar nutrient concentrations of slash pine in the first 11.5 years to the initial fertilization and the additional P applied at age 10 years.Foliar N and K concentrations in the first 9.6 years of plantation decreased with the initial P rate. Application of the basal fertilizers improved foliar Cu concentration. Foliar Ca and Mg concentrations increased linearly with the initial P rate. The initial fertilization did not affect foliar Mn concentration in the first 9.6 years. Foliar P concentration increased quadratically with the initial P rate, which accounted for 77–86% of the variation in foliar P concentration. Most of the explained variation in foliar nutrient concentrations was attributable to the plantation age except for foliar P concentration. In the case of foliar P concentration, 53% was explained by the initial P rate, 31% by the plantation age and 2% by the positive interaction between the initial P rate and the plantation age. Foliar P concentration of slash pine at age 11.5 years increased quadratically with the initial P rate and linearly with the additional 40 kg P ha–1 applied at age 10 years, accounting for 81% of the variation in the foliar P concentration. A mathematical model has been proposed for predicting the changes in soil nitrogen status due to continuous fertilization in a continuous cropping sequence. The model also enabled the prediction of the steady state of soil nitrogen for a specified fertilizer practice.The model was applied to six years nitrogen availability data of four fertilizer practices in finger millet-maize-cowpea sequence followed in the Long Term Fertilizer Experiments conducted at Tamil Nadu Agricultural University, Coimbatore, India. Dairy farming is the main agricultural activity of the Basque Country. A dairy farm is characterized as a system with soils and crops, forage, cattle and manure as main components, and in such a system, nutrient cycling is very important to maintain soil fertility and optimize forage production. To quantify nutrient transfers in the cycle, a simple system was developed and has been applied to seventeen farms to examine its ability to achieve a balanced P and K fertilization. These farms have provided data on inputs (fertilizer, feeds, concentrates), pasture and manure management, and outputs (milk production), and soil samples have been taken from farm pastures. Phosphorus and K in excreta and uneaten pasture is used with a relatively high efficiency as suggested by the relatively high efficiency of P and K utilization by the pasture that usually ranges from 70 to 90%. Concentrate feeding (3000 kg cow–1 yr–1) represents one of the main P and K inputs in Basque Country dairy farms, averaging 26 and 66 kg ha–1, respectively. Besides, release of K in the soil through slow liberation from non-exchangeable sites was estimated as 30 kg ha–1. Thus, a high efficiency in excreta recycling would diminish substantially P and K mineral fertilizer needs. Farm nutrient budgets appear to be a convenient tool for determining nutrient shortages and surpluses at farm level, and thus they are considered as a first step to support a better management of maintenance fertilization of permanent pastures. 相似文献

Nitrogen demand from soybean seeds during seed filling is very high and has been proposed as the cause of nitrogen remobilization and leaf senescence. Previous research has not shown consistent effects of late season fertilization on seed yield, while its effects on leaf senescence have not been evaluated. Two field experiments were performed to determine the effects of a late season N fertilization on leaf senescence and fall, seed yield and its components, and residual soil nitrate, and to evaluate the potential risk of groundwater contamination. Two rates of nitrogen (50 and 100 kg N ha–1) were applied at the R3 and R5 development stages. Nitrogen fertilization, either at R3 or R5, increased soil nitrate availability during the seed-filling period. Seed yield, seed number and protein content were not affected by N fertilization. The addition of 100 kg N ha–1 produced a small delay of 1–2 days in the leaf fall, and slightly increased seed size (3.6%). Our results suggest that increasing soil N availability during the seed-filling period is not an effective way to delay leaf senescence or to increase seed growth and yield of soybean. Nitrogen fertilization increased the level of residual nitrate in the top soil at one site (the one with lowest seed yield), increasing the risk of nitrate leaching during subsequent fallow. 相似文献
