Two sets of traditional Greek sheep milk yoghurt were produced: the first one (YC) using normal yoghurt culture (Lactobacillus delbrueckii subsp. bulgaricus ?10.13 and Streptococcus thermophilus ?10.7) and the second (PR) with the same normal culture mixed with Lactobacillus paracasei subsp. paracasei DC412. YC and PR had similar physicochemical properties and proteolysis patterns throughout storage. Both products showed similar peptide profiles by RP-HPLC but quantitative differences were observed in respect to storage time. Single-strain cultures of the microorganisms used showed similar peptide profiles for both lactobacilli, yet L. delbrueckii subsp. bulgaricus was the most proteolytic of all three microorganisms. The peptide content and the ACE-inhibitory activity of the water-soluble extracts of yoghurts, YC and PR, increased throughout storage. Major peptides were identified from yoghurt PR and from the separate cultures of L. delbrueckii subsp. bulgaricus and L. paracasei subsp. paracasei. Most of these peptides were derived from β-casein. A peptide, β-CN f114-121, with well-established ACE-inhibitory and opiate-like activity was identified in yoghurt PR. Further identified peptides were regarded as potential ACE-inhibitors according to their sequence.  相似文献

In the present study, 122 food samples from the German food market were analysed for their C18:1 trans fatty acid (TFA) content and profile. A particular focus of the survey were baked and fried foods. TFA analysis was performed by means of silver ion SPE (Ag+‐SPE) in combination with high‐resolution GC (HRGC‐FID). Overall, 51 bakery product samples were analysed of which 25 samples were prepacked bakery products purchased from local retail stores and 26 samples of unpacked bakery products purchased from local bakery shops. In addition, 14 French fries samples obtained from small local fast food restaurants as well as from internationally operating fast food chains, 27 potato and tortillas chips, 15 instant soups as well as 15 dry culinary sauces were analysed. The highest amounts of C18:1 TFA isomers were found in deep‐fried bakery products. Prepacked branded cookies and biscuits on the other hand contained only negligible C18:1 TFA amounts. Regarding their C18:1 trans isomer profile most deep‐fried bakery products exhibited a Gaussian‐distributed isomer profile. The analysed prepacked croissants, cookies and biscuits contained predominantly ruminant TFA (TFA) as suggested by the presence of vaccenic acid (C18:1 trans 11), which was the major C18:1 TFA isomer in these products. All non‐bakery samples (n = 71) contained less than 3 g C18:1 TFA per 100 g fat. In conclusion, TFA still occur in considerable amounts in a few German food products, especially in some deep‐fried bakery products (‘Berliner’ type of doughnuts). Practical applications: Trans fatty acids, in particular the trans octadecenoic fatty acid isomers (C18:1), are generally considered from the nutritional point of view as undesirable food components due to their negative health effects. Tremendous efforts have been made by major food processors in order to decrease or even eliminate the presence of TFA in some foodstuffs (e.g. in margarines in European countries). However, some food processors of other food sectors are still applying oils and fats containing partially hydrogenated vegetable oils, whereas others within the same food category have already switched their processing conditions and/or raw materials towards TFA alternatives. Therefore, actual TFA data of foodstuffs determined by means of state‐of‐the‐art analytical procedures (Ag+‐SPE in combination with GC‐FID) is necessary to detect areas of further improvement in the food supply chain and to provide data for an update of dietary TFA intake.  相似文献

The total quantity (11.7 mg/kg of pulp) of aglycones released by enzymatic hydrolysis of the glycosidic extract indicates a significant aroma potential for cupuacu. Among the 47 aglycones identified, 24 are not present in the free volatile fraction, among them, 4-methylguaiacol, 4-propylguaiacol, 2,6-dimethyl-octa-1,7-dien-3,6-diol, 2,6-dimethyl-oct-7-en-1,6-diol, homovanillic acid, 2-methyl-but-3-en-1-ol and acetic acid. The quantitatively most important aglycones are 3-methyl-butan-1-ol, 2-phenylethanol, linalol, (Z)-2,6-dimethyl-octa-2,7-dien-1,6-diol, butan-1-ol and hexan-1-ol. The methylated alditol acetates analysis of monosaccharides released by acid hydrolysis of the heterosidic extract, indicates that glucose is involved in glucosidic and glycosidic structures. Moreover, rhamnopyranose and xylopyranose units, bound in the terminal position, suggest the presence of rutinosides and small amounts of primeverosides. Six glucosides, hexyl, benzyl, 2-phenylethyl, (R) and (S)-linalyl and geranyl β- -glucopyranosides, five rutinosides, benzyl, (R) and (S)-linalyl, -terpineyl and 2-phenylethyl rutinosides and 3-methyl-but-2-enyl vicianoside were identified by GC–EIMS after TFA derivatization of the crude heterosidic fraction. Moreover, four linalol oxides, octyl, 3-methyl-butyl β- -glucopyranosides and hexyl, octyl, two linalol oxides and 3-methyl-butyl rutinosides, have been tentatively identified from their TFA mass spectra. Some glucosides are probably substituted by malonyl and unidentified acyl residues.  相似文献

This paper describes the influence of slaughter season on lipid content, fatty acid composition, conjugated linoleic acid (CLA) isomeric profile and nutritional value of fat in Barrosã veal from calves reared according to the specifications of the Protected Designation of Origin (PDO). Barrosã purebred calves (n = 27) were raised in a traditional production system and slaughtered in early autumn (October) and late spring (June). Barrosã-PDO veal only presented seasonal differences in the levels of some minor fatty acids and CLA isomers, as well as in the PUFA/SFA ratio. Based on the analysed grass intake indicators, it was shown that veal-PDO has similar values to pasture-fed cattle for both slaughter seasons. From a human nutrition perspective, intramuscular fat in Barrosã-PDO veal has a high nutritional value throughout the year, since CLA contents and the percentages of the c9,t11 isomer are relatively high, and the n − 6/n − 3 ratios are within the recommended values for the human diet.  相似文献

Inhibition of angiotensin I-converting enzyme by wheat gliadin hydrolysates   总被引:1,自引:0,他引:1  
A tryptic gliadin hydrolysate was fractionated into peptide fractions, which were assigned to either the central domain (CD) or terminal domains (TD) of gliadins. The domains were expected to contain amino acid (AA) sequences which, when released from the parent protein, inhibit the angiotensin I-converting enzyme (ACE), which plays a key role in regulating blood pressure. A proline (Pro) poor TD related fraction, containing the smallest peptides, showed the highest ACE inhibitory activity (IC50 = 0.33 mg/ml). Additional peptidases were selected based on their in silico predicted ability to release ACE inhibitory peptides. Further hydrolysis of the tryptic hydrolysate fractions with thermolysin, Clarex, Alcalase and Esperase increased ACE inhibitory activities. Immobilised Ni2+-ion affinity chromatography (IMAC) purification of a TD related peptide fraction obtained by sequential hydrolysis with trypsin and thermolysin yielded a fraction with an IC50 value of 0.02 mg/ml. This IMAC fraction was enriched in histidine and hydrophobic AA (Pro, Val, Ile, Leu and Phe).  相似文献

当管道同时发生多个泄漏点时,多个泄漏点的声波信号出现混叠,从而影响泄漏声信号的传播和衰减规律。在多点泄漏声波信号混叠的情况下,传统的泄漏定位方法无法对多个泄漏点进行有效定位。为此,提出了一种基于时频分析和改进差分进化的多点泄漏定位方法。由于变分模态分解(variational mode decomposing, VMD)的分解模态个数对特征提取有影响,因此采用能量函数估计分解模态个数,从而对VMD改进。然后采用改进VMD对多点泄漏声波信号进行处理。另外,通过时频分析(time-frequency analysis, TFA)估计多个泄漏点数量,从而得到多点泄漏的定位函数。差分进化(differential evolution, DE)算法易陷入局部最优,在进化后期收敛速度较慢,采用粒子群优化(particle swarm optimization, PSO)算法提高DE的全局收敛速度,并通过改进的差分进化算法估计多个泄漏点的位置。在管道现场进行多点泄漏的模拟试验,试验结果表明该方法能够准确地估计多点泄漏位置,泄漏定位的最小误差为18 m。  相似文献

Acetic acid, lactic acid, and trifluoroacetic acid were used as complex agents for the preparation of dense barium zirconate fibers with electrospinning method. The formation of lactate when lactic acid was used as complex agent led to a mild decomposition process which was beneficial for the fabrication of dense barium zirconate fibers. High temperature stable intermediate phase of BaF2 appeared when trifluoroacetic acid was used. The intermediate phase of BaF2 was disadvantageous for pure and dense barium zirconate formation. While water vapor preheat treatment for BaF2 contained fibers was an effective way for the fabrication of pure and dense barium zirconate fibers.  相似文献

针对脚手架坍塌事故在工程项目中发生比较频繁,造成的后果比较严重。通过运用贝叶斯网络的故障树方法,不仅避免了传统故障树法的缺点,也定量找出脚手架坍塌事故的主要因素。首先应用故障树法建立风险因素之间的逻辑关系,并按照转化规则将故障树模型转化成贝叶斯网络模型,应用贝叶斯网络的推理能力计算出脚手架坍塌的先验概率以及各底事件的后验概率。另外,一个事件不能仅仅用发生和不发生描述,所以将部分基本事件由二态修正成三态,计算出先验概率和后验概率。通过以上得到的结果进行对比分析,从而表明,考虑事件的多态性可以更加准确的得到事件的概率分布。最后针对脚手架中发生后验概率比较大的事件,提出一些预防脚手架坍塌的措施。  相似文献

A large number of polysaccharides present in the leaves of Taxus chinensis var. mairei were successively extracted with hot water after preliminary treatments. The ion-exchange and gel-permeation chromatography was used to isolate and purify the major polysaccharides to afford a complex water-soluble polysaccharide, named T1 with a molecular mass of 3.44 × 10Da determined by HPGPC. T1 consisted of 2,4-di-O-methyl-mannose, rhamnose, arabinose, xylose, galactose, mannose, galacturonic acid and glucuronic acid in a molar ratio of 2:5:24:9:3:46:1:10, and the acetyl content was estimated to be 2.1%. On the basis of methylation analysis, periodate and chromium trioxide oxidations, Smith degradation, graded acid hydrolysis, and NMR and IR spectroscopy, T1 possessed a 1,3,6-linked β-d-Manp main chain, substituted by mannose, arabinose, galactose, xylose, rhamnose, 2,4-di-O-methyl-mannose, galacturonic acid, and glucuronic acid residues. These contained non-reducing end-units of mannose, arabinose, xylose, 2,4-di-O-methyl-mannose, and glucuronic acid. This is the first report on isolation of an acid heteropolysaccharide from Taxus species.  相似文献
