藻胆蛋白免疫荧光法的初步探索

摘    要:采用CNBr活化的琼脂糖凝胶小珠作为固相载体,应用藻胆蛋白荧光标记物对血清可溶性抗原检测作了初步的探索. 结果表明, 藻胆蛋白免疫荧光法用于可溶性抗原检测不但可行,而且具有荧光强度高、可长期保存且无明显衰减等特点.

Abstract:Phycobiliprotein fluorescence probe was used in preliminary experiment of serum soluble antigen detection. The immunological system was designed by means of solid phase sandwich technique in diagnosis of Hepatitis B surface antigen. CNBr-activated Sepharose 4B was used to prepare capture antibody coating microsphere. R-phycoerythrin labeled antibody was added to trace the positive serum sample. The result showed that the feasibility was proved. Positive sample turned out to be bright beads with orange fluorescence under fluorescence microscope, while the negative one was apparently different in its color with little orange fluorescence. And also, the fluorescence of diagnosis result can be preserved for a long time with little attenuation of fluorescence intensity.
