超声法提取松针叶绿素及稳定性研究

摘    要:以白皮松为原料,使用乙醇作提取剂,采用超声法对松针叶绿素的提取方法进行了研究.通过单因素实验和正交试验分别考察了提取时间、乙醇浓度、体积料液比、溶液温度对提取率的影响.得到超声法最佳提取条件为无水乙醇作提取剂,体积料液比1∶4,溶液温度40℃,超声时间15min.在此条件下的提取率为94.4%.叶绿素铜钠盐的制备:以松针为原料,将含Mg离子的叶绿素通过置换反应制得水溶性的叶绿素铜钠盐,再用乙醇浸提,然后用石油醚分离出脂溶性成份,最后制得叶绿素铜钠盐.结果表明,叶绿素铜钠盐在常温下可以存放30d,叶绿素的稳定性大为提高.

Abstract:The supersonic extraction method of chlorophyll was investigated using pinus lacebark pine as raw material, with ethanol as extraction solvent. By the single fachor and orthogonal design, the impact of extraction rate was investigated at the exist of these factors:time for supersonic extraction, concentration of ethanol, solidsolvent volume ratio and temperature of the solution. are:anhydrous ethanol as ethanol as extraction advent The optimized extraction conditions of supersonic method solid-solvent volume ratio 1 : 4 in 40℃ of 15rain. The extraction rate was 94. 4%. Preparation of the sodium copper chlorophyllin: chlorophyll with Mg was tuned into water solable sodium copper chlorophyllin by replacement reaction using needles as raw material, with ethonal as extraction solvent, using petroleum separate liposoluble part to gain the sodium copper chlorophyUin was investigated. The result showed that the sodium copper chlorophyllin can be preserved in nomal temperature for 30 days and stability of chlorophy was improved greatly.
