合欢花中总黄酮的提取工艺及对羟自由基清除作用的研究

摘    要:
采用乙醇为浸提溶剂从合欢花中提取黄酮类化合物,采用分光光度法测定提取液中黄酮类化合物的含量,通过单因素实验考察了乙醇浓度、料液比、浸提时间、浸提温度对合欢花总黄酮提取率的影响,在单因素实验基础上确定正交实验因素及水平,对提取条件进行优化,并且研究了合欢花提取液中黄酮类化合物对.OH的清除作用。实验结果表明,各因素对合欢花总黄酮提取率的影响程度依次为:乙醇浓度>料液比>浸提温度>浸提时间;提取的最佳工艺条件为:乙醇浓度50%,料液比1∶25,提取时间2h,浸提温度70℃,在此条件下,合欢花总黄酮提取率为2.53%;合欢花中总黄酮提取液对.OH自由基有一定的清除作用,当合欢花总黄酮浓度达到0.06mg/mL时,对.OH自由基清除率为62.81%。

Abstract:
The flavonoids from flower of silktree albizzia was extracted by ehtanol.The total flavonoids in the extracts were measured by spectrophotometer.The effect of ethanol concentration,solid to liquid ratio,extraction time,extraction temperature on the extraction ratio of were investigated by single factor and orthogonal experiment.Its hydroxyl free radical scavenging ability was evaluated.The results showed that the order of significant factors that affected on the extraction ratio was as follows:ethanol concentration>solid to liquid ratio>extraction temperature >extraction time.The optimum extraction technology was established as follows:50% ethanol solution as solvent,solid to liquid ratio 1 ∶25,extracting temperature 70℃ and extracting time 2h.Under the above condition,the extraction ratio of flavonoids was 2.53%.Total flavonoids from flower of silktree albizzia had effects on scavenging of hydroxyl radical.Scavenging rate of flavonoids on hydroxyl free radical was 62.81% when flavonoids concentration was 0.06mg/mL.
