化妆品中天然红4含量的检测方法研究

摘    要:建立了一种化妆品中天然红4含量的高效液相色谱紫外检测方法。样品采用盐酸在沸水浴下提取,在酸性条件下用聚酰胺固相萃取小柱富集、净化,氨水/甲醇溶液洗脱后检测;色谱条件为:XB-C18色谱柱(5μm,4.6 mm×150 mm),以φ=0.3%的甲酸溶液和乙腈为流动相,流速为1.0 mL.min-1,检测波长为276 nm。结果表明,在此条件下,天然红4(以胭脂红酸计)在0.255~5.10 mg.L-1范围内呈良好的线性关系(相关系数r=0.999 7)。天然红4检测限为0.2μg.g-1,平均回收率为87.4%~110.8%,RSD为3.4%~8.6%。

Abstract:A method for determination of natural red 4 in cosmetics using high performance liquid chromatography-ultraviolet visible detector(HPLC-UV) was estabilished.The sample was extracted by hydrochloric acid solution in boiling water bath,purified and enriched by a polyamide SPE cartridge under acidic condition,then eluted by ammonia/methanol solution and measured by HPLC-UV.The chromatographic separation was achieved with an XB-C18 column(5 μm,4.6 mm×150 mm) by using 0.3% formic acid and acetonitrile as mobile phase with a flow rate of 1.0 mL·min-1.The detection wavelength is 276 nm.The correlation coefficient(r) of natural red 4(based on carminic acid) is 0.999 7 in the linear range of 0.255~5.10 mg·L-1.The detection limit of natural red 4 is 0.2 μg·g-1.The average recovery ranges from 87.4% to 110.8%.The RSD is in the range of 3.4% to 8.6%.
