白介素-2佐剂狂犬病疫苗的免疫学效果

目的研究白介素-2佐剂狂犬病疫苗的免疫学效果。方法将人用纯化Vero细胞狂犬病疫苗原液与基因工程白介素-2按一定比例混合,制成白介素-2佐剂狂犬病疫苗,另将疫苗原液加氢氧化铝,制成铝佐剂狂犬病疫苗。将白介素-2佐剂疫苗、氢氧化铝佐剂疫苗和无佐剂疫苗分别于0、7d经腹腔免疫昆明小鼠,0.5ml/只,并在免疫前和免疫后4、6、10、17、2、31、45和59d,眶静脉采血,用快速免疫荧光灶抑制试验检测小鼠血清中和抗体。同时于0、7d经腹腔免疫BALB/c小鼠,0.5ml/只,第15天取脾检测淋巴细胞转化率。结果白介素-2佐剂疫苗与氢氧化铝佐剂疫苗和无佐剂疫苗淋巴细胞转化率差异均有显著意义。与铝佐剂疫苗和无佐剂疫苗相比,白介素-2佐剂疫苗诱导产生中和抗体的时间早,抗体滴度高。结论白介素-2佐剂狂犬病疫苗具有良好的免疫学效果。

Immune Effect of Rabies Vaccine Using Interleukin-2 as Adjuvant

Objective To study the immune effect of rabies vaccine using interleukin-2(IL-2)as adjuvant.Methods Mix the bulk of purified rabies vaccine(Vero cells)for human use with recombinant IL-2 according to a certain ratio to prepared the rabies vaccine containing IL-2 adjuvant.Meanwhile,mix the bulk with aluminium hydroxide to prepare rabies vaccine containing aluminium hydroxide adjuvant.Prepare adjuvant-free rabies vaccine with the bulk of same batch.Perform abnormal toxicity and potency tests on the three kinds of vaccines.Divide Kunming mice into three groups and immunize i.p.with the three kinds of rabies vaccines on days 0 and 7 respectively,0.5 ml per mouse.Collect blood sample before immunization and on days 4,6,10,17,31,45 and 59 respectively for de- termination of serum neutralizing antibody by rapid immunofluorescence focus inhibition test(RIFIT).Divide BALB/c mice into three groups and immunize i.p.with the three kinds of rabies vaccines on days 0 and 7 respectively,0.5 ml per mouse,then determine the lymphocyte transformation rate on day 15.Results All the rabies vaccines were qualified in abnormal toxicity test.The potency of rabi- es vaccine containing IL-2 adjuvant showed no significant difference with that of vaccine containing aluminium hydroxide,but was signif- icantly higher than that of adjuvant-free vaccine.The lymphocyte transformation rate induced by rabies vaccine containing IL-2 adjuvant was significantly higher than those induced by adjuvant-free vaccine and the vaccine containing aluminium hydroxide adjuvant.Com- pared with adjuvant-free vaccine and the vaccine containing aluminium hydroxide adjuvant,the rabies vaccine containing IL-2 adjuvant induced neutralizing antibody early,and the antibody titer was high.Conclusion The rabies vaccine using IL-2 as adjuvant showed good immune effect.
