摘    要:以金针菇为试验材料,对液体种子发酵培养基的组分进行了筛选及优化。 以pH值、还原糖、氨基酸态氮、酶活力、菌种生物量 和菌丝球数目为评价指标,选择碳源、氮源、无机盐、维生素为影响因素,通过正交试验确定液体培养基配方。 研究结果表明,金针菇 的最佳液体培养基配方为:可溶性淀粉3%、黄豆粉5%、KH2PO4 0.05%、VB2 0.005 0%。 在此优化条件下,蛋白酶、纤维素酶和淀粉酶活 力最高,分别为1.28 U/mL、1.25 U/mL和1.84 U/mL,相应的菌丝干质量为0.71 g/100 mL。

Abstract:Taking Flammulina velutipes as the tested material, the composition of liquid seed fermentation medium was screened and optimized. Using pH value, reducing sugar, amino acid nitrogen, enzyme activity, bacterial biomass and the number of mycelium pellet as evaluation indexes, and car- bon source, nitrogen source, inorganic salt and vitamin as evaluation factors, the liquid medium formula was optimized by orthogonal tests. The re- sults showed that the optimum formula of medium for F. velutipes was as follows: soluble starch 3%, soybean bean flour 5%, KH2PO4 0.05%, VB2 0.005 0%. Under these conditions, the protease activity, cellulase activity and amylase activity were the highest, which were 1.28 U/ml, 1.25 U/ml and 1.84 U/ml, respectively. The corresponding dry weight of mycelium was 0.71 g/100 ml.
