摘    要:为提高汉逊氏葡糖酸醋杆菌(Gluconacetobacter hasenii)JR-02细菌纤维素(BC)合成能力,采用响应面法对其发酵工艺参数进行优化。通过单因素试验和Plackett-Burman试验筛选出对菌株JR-02的细菌纤维素产量影响较为显著的4个因素为初始pH、MgSO4、玉米浆干粉、乙醇,再通过Box-Behnken试验设计最终确定最佳发酵工艺为:葡萄糖30 g/L、玉米浆干粉15 g/L、乙酸3 g/L、Na2HPO4 3 g/L、MgSO4 0.11 g/L、CaCO3 0.8 g/L、乙醇9 mL/L、初始pH 4.5、30 ℃、160 r/min。优化后BC产量达到(4.74±0.15) g/L,为未优化前BC产量的1.87倍。

Abstract:The fermentation process parameters were optimized by response surface method for improving the bacterial cellulose (BC) synthesis capacity of Gluconacetobacter hasenii JR-02. Four factors which had significant effect on bacterial cellulose yield were determined as initial pH, MgSO4, corn steep powder and ethanol by single factor and Plackett-Burman experiments. The optimum fermentation process was determined as glucose 30 g/L, corn steep powder 15 g/L, acetic acid 3 g/L, Na2HPO4 3 g/L, MgSO4 0.11 g/L, CaCO3 0.8 g/L, ethanol 9 ml/L, initial pH 4.5, culturing at 30 ℃ and 160 r/min. After optimization, bacterial cellulose yield was up to (4.74±0.15) g/L, which was 1.87 times higher than that of before optimization.
