Apoptosis induced by LED- activated Hypocerllin B in human ovariancarcinoma cells using fluorescent staining analysis

Abstract:Objective: To observe the photochemical biological effect of Lighting Emitting Diode(LED) - activated Hypocerllin B on the apoptosis of ovarian carcinoma cells. Methods: A special lighting emitting diode equipment was employed to investigate the photochemical biological effect of Hypocerllin B 4h after photedynamic therapy in a human ovarian carcinoma cell line H08910 by Hoechst33258 fluorescent staining analysis. Results: Hoechst33258 staining showed that the morphological changes in the nuclear ehromatin of LED- activated Hyvx~rllin B in H08910 were extraordinarily siofificant.ln the control groups, the nuclei were stained a less bright blue and the color was homogeneous. After being irradiated by LED with Hypocernin B for 4h, the blue emission light in apoptotic cells was much bright than the control cells. Condensation of ehromatin and nuclear fragmentations could also be found in many treated cells and some of them formed the structure of apoptotic bodies, which is one of the classic characteristics of apoptotie cells. Conclusion: Lighting Emitting Diode (LED) - activated Hypocerllin B could effectively induce apoptosis of human ovarian carcinoma cells and Lighting Emitting Diode(LED) will become an alternative light source for photedynamic therapy.
