表面活性剂作为糖蜜发酵谷氨酸促发剂的研究

摘    要:通过对预先选择的5种表面活性剂作为谷氨酸发酵促进剂效果的初步筛选,得知五甲基三聚氧乙烯三胺和3-氯-2-羟丙基三乙基氯化铵对提高产酸率有较为明显的效果;进而从加入时间、加入剂量两个方面研究了筛选出的两种表面活性剂对谷氨酸棒状杆菌(Corynebacterium glutamicum)T-613菌体生长和谷氨酸发酵产酸的影响,从而得到最佳的发酵工艺参数。研究结果表明:发酵中后期(22 h)加入质量分数为0.5%3-氯-2-羟丙基三乙基氯化铵0.8 mL,菌种生长对数期(9 h)加入质量分数为1%的五甲基三聚氧乙烯三胺型表面活性剂2 mL,在不影响菌种生长的前提下,促使谷氨酸发酵产酸率由0.8718%提高到1.8027%,有效地提高了生产效率。

Abstract:According to comparing the effect of 5 pre-selective surfactants on glutamic acid fermentation,3-chloro-2-hydroxypropyl trethyl ammonium chloride and pentamethyl tripolyoxyethylene triamine can improve the rate of the glutamic acid production(RG) obviously.Then the effect of the two kinds of surfactants on cell growth and glutamate acid production by Corynebacterium glutamicum T-613 was investigated in the respects of additive time and additive dosage.Experimental results showed that adding 1.25 %pentamethyl tripolyoxyethylene triamine 0.3 mL in 22 h since the beginning of the fermentation and 0.3 % 3-chloro-2-hydroxypropyl trihyl ammonium chloride 2.5 mL in 9h at the stain growth logarithmic phase: Under the condition of stain growing normally,the product yield rose from 0.8718 % to 1.8027 %.
