摘    要:通过选择检测波长和优化色谱条件,建立一种8 min内分离葡萄酒中13种酚酸的反相超高效液相色谱(UPLC)检测方法。色谱条件:以2%乙酸和纯甲醇为流动相,采用ACQUITY UPLC BEH C18(100 mm×2.1 mm,1.7 μm)色谱柱,流速为0.30 mL/min,检测波长为280 nm和320 nm,柱温为50 ℃,进样量为1 μL。结果表明:13种酚酸的最低检测限均<0.10 mg/L,标准曲线线性相关性(R2)均>0.99,回收率为90%~110%,且其相对标准偏差(RSD)均<3.00%,保留时间及峰面积的RSD均分别<1.00%、3.00%。该方法灵敏度高、速度快、分离度高,精密度及稳定性良好。样品中检测结果表明,该方法适合葡萄酒中酚酸的检测。

Abstract:By selecting wavelength and optimizing chromatographic condition, a rapid determination method for separating 13 phenolic acids in wine in 8 min was established using reversed-phase ultra performance liquid chromatography (RP-UPLC). 13 phenolic acids were separated by ACQUITY UPLC BEH C18 (100 mm×2.1 mm, 1.7 μm) with mobile phases of 2% acetic acid and pure methanol. The chromatographic conditions was as follows: flow rate 0.30 ml/min, the UV detection wavelength 280 nm and 320 nm, column temperature 50 ℃, injection volume 1 μl. The results indicated that the lowest detectable limit of 13 phenolic acids was less than 0.10 mg/L, the correlation coefficients (R2) was more than 0.99. The recovery rates were in the range of 90%-110% and the relative standard deviation (RSD) was less than 3.00%. The RSD values of retention time and peak area were less than 1.00% and 3.00%, respectively. Due to its high sensitivity, fast speed, high separation, good precision and reliable stability, the method was suitable for the detection of phenolic acids in wine.
