摘    要:以谷氨酸棒杆菌ATCC13032为出发菌,研究了青霉素浓度及溶菌酶浓度对原生质体形成率的影响,以及氯化钙和二甲基亚砜等物质的添加对其再生率的影响.结果表明最佳青霉素浓度及溶菌酶浓度分别为0.6 U/mL和1 mg/mL,在此条件下谷氨酸棒杆菌ATCC13032原生质体形成率达96%.为进一步提高原生质体再生率,尝试了4种添加物,其中氯化钙及二甲基亚砜能显著提高原生质体再生率.当3 g/L氯化钙和10 mL/L的二甲基亚砜组合添加时原生质体再生率更可达34%,是未添加的2.1倍,达到采用夹层法培养能获得再生率的最大值.

Abstract:The strain corynebacterium glutamicum ATCC 13032,a typical corynebacterium glutamicum strain,was used to prepare protoplast.Penicillin concentration,lysozyme concentration and additives were examined systematically.When penicillin concentration,lysozyme concentration were 0.6U/mL and 1 mg/mL,the rate of obtained protoplasts was 96%.For enhancing the regeneration frequency of the protoplast,4 kinds of additives were applied.Within the scope studied,both CaCl2 and DMSO were shown to be effective.The most pref...
