绿色木霉HZ012发酵生产几丁质酶的研究

摘    要:为探讨绿色木霉发酵合成几丁质酶的最优化培养条件,测定了各种不同培养条件下绿色木霉发酵合成几丁质酶的活力。结果表明:用0.9%几丁质和3.0%葡萄糖作为双碳源时几丁质酶的产量最高达0.192U/mL,高于单独使用葡萄糖或几丁质作为碳源时的最高产量;以1.0%蛋白胨作为氮源时几丁质酶的最高产量高于用1.0%酵母膏、1.0%硫酸铵、1.0%牛肉膏和1.0%亚硝酸钠作为氮源时的最高产量;发酵液的最佳初始pH为5.0,最佳微量元素添加量为2mL/L;在上述最优化培养条件下培养44h时几丁质酶活力达到最高值0.248U/mL。该方法获得的几丁质酶活力与Kapat等人报道的用Trichodermaharzianum生产几丁质酶的结果相比,酶活提高33%以上。

Abstract:In order to explore the optimal culture conditions for the synthesis of chitinase from Trichoderma viride HZ012, the activity of chitinase was detected in different culture conditions. The results indicated that the highest activity of chitinase was 0.192 U/mL using 0.9% chitin and 3.0% glucose as double carbons source in the medium, which was higher than those using chitin or glucose as the sole carbon source. The activity of chitinase using 1.0% peptone was higher than those using 1.0% yeast extract, 1.0% ammonia sulfate, 1.0% beef extract and 1.0% sosodium nitrite as nitrogen source, respectively. The highest activity of chitinase of 0.248 U/mL was obtained at 44 h in the optimal medium with the initial pH 5.0 and the trace elements solution additive amount 2 mL/L. Compared with that of chitinase by Trichoderma harzianum reported by Kapat, the activity of chitinase obtained under the experimental conditions was higher than 33%.
