过量积累γ-亚麻酸的深黄被孢霉突变株MI-33的发酵研究

摘    要:对过量积累γ-亚麻酸的深黄被孢霉突变株MI-33的发酵条件作了初步研究。通过试验不同接种方式对发酵的影响,确定了接种菌丝体为合理的接种方式;通过试验不同碳源对发酵的影响,确定了葡萄糖为产生γ-亚麻酸的最适碳源;菌体生长动态的研究表明,合适的种子液培养时间为48h;发酵动态的研究表明,合适的发酵终止时间为96h,此时干菌体收率为26.51g/L、油脂产率为14.08g/L、GLA产率为901.16mg/L。

Abstract:The fermentation of Mortierella isabellina mutant MI-33 overproducing gamma-linonenic acid(GLA) was preliminarily studied. Suitable way of inoculation was determined to be inoculating with mycelia by studying the effects of different inoculation ways on fermentation. Suitable carbon source for GLA accumulation was determined to be glucose by studying the effects of different carbon sources on fermentation. The dynamics of mycelial growth showed the optimal age of seed culture was 48h. The dynamics of fermentation showed the optimal age of fermentation for GLA production was 96h, when dry cell weight, lipid yield and GLA yield reached 26.51g/L, 14.08g/L and 901.16mg/L, respectively.
