摘    要:该研究以小麦胚芽为原料,2,6-二甲氧基对苯醌(2,6-DMBQ)产量为评价指标,对发酵菌种进行筛选并考察原料预处理方式的影响。在此基础上,采用单因素试验及正交试验对发酵条件进行优化。结果表明,最适发酵菌种为低糖面包酵母;最佳原料预处理方式为超声处理;低糖面包酵母发酵小麦胚芽产2,6-二甲氧基对苯醌的最优发酵条件为菌种添加量1∶8(酵母与麦胚质量比),料液比1∶15(麦胚与水质量比),发酵时间42 h,发酵温度28 ℃。在此最优发酵条件下,2,6-二甲氧基对苯醌的产量最高为(765.23±9.11) μg/g。

Abstract:The culture system of Saccharomyces cerevisiae was optimized by response surface methodology, in order to improve the fermentation biomass of S. cerevisiae in industrial production. According to the single-factor experiments, results showed that the optimal condition for S. cerevisiae culture system was starch sugar as carbon source, corn pulp and urea as nitrogen source, K2HPO4 as buffer salt and MgSO4 as trace elements, and the optimal vitamin B group was VB2 , VB3 and VB8 . The main factors affecting the biomass of S. cerevisiae were starch sugar, corn pulp and urea through the Plackett-Burman design and the peak range of the main factors were obtained by the steep climb path experiments. Box-Behnken design and response surface methodology were used to optimize the main factors. The results showed the optimal fermentation conditions were starch sugar 28.8 g/100 ml, corn pulp 6.1 g/100 ml, urea 0.097 5 g/100 ml, K2HPO4 0.1 g/100 ml, MgSO4 0.05 g/100 ml, vitamin B2 0.1 mg/100 ml, vitamin B3 0.1 mg/100 ml, and vitamin B8 0.1 mg/100 ml. Under the optimized cultivation system, the OD560 nm value of S. cerevisiae was 60.60, which was 5.27 times as much as that of by yeast extract peptone dextrose medium.
