摘    要: 以HVI500矿物基础油为唯一碳源进行选择性富集培养,从石油污染土壤中筛选出1株菌,命名为HVI0901。采用CODCr法测定了含HVI500矿物基础油培养液的COD值,由于HVI500矿物基础油被降解而使培养液的COD值降低,确定HVI0901为HVI500矿物基础油降解菌。通过形态学观察、生理生化实验和16S rDNA基因序列比对分析,初步确定HVI0901属于戈登氏菌属(Gordonia)。采用倾注平板法计数不同温度条件下HVI0901的菌落,用CODCr法测定不同pH值条件下HVI500矿物基础油培养液的COD值,探讨了温度和pH值对HVI0901降解基础油的影响。结果表明,HVI0901在不同温度和pH值条件下的降解能力不同,在32℃、pH值为7.8时,其降解HVI500矿物基础油的作用最佳。

Abstract:By using HVI500 mineral base oil as sole carbon source,a bacterial strain,named as HVI0901, was isolated from the petroleum-contaminated soil through selective enriched culture. The COD value of the culture fluid with HVI500 mineral base oil was determined by the CODCr method. Result showed that COD value decreased because HVI500 mineral base oil was degraded,indicating that the strain HVI0901 was a degrading bacterium for HVI500 mineral base oil. The strain HVI0901 was preliminarily identified as genus Gordonia through morphological observations, physiological and biochemical tests and 16S rDNA sequencing analysis. The impacts of the strain under different temperatures and pH values on biodegradability of HVI500 mineral base oil were evaluated by the pour plate count method and the CODCr method,respectively. Results demonstrated that the biodegradation abilities of HVI0901 under different temperatures and pH values were different, and the optimum biodegradation temperature and pH value for HVI0901 were 32 ℃ and 7.8, respectively.
