葡萄籽中活性成分提取工艺的研究

摘    要:采用V (丙酮 )∶V (水 ) =2∶3和V(乙酸乙酯 )∶V(水 ) =8 5∶1 5两种溶剂体系从葡萄籽中提取活性成分。对比分析了两种体系的优劣 ,结果表明 :丙酮 -水体系提取过程复杂 ,提取率为0 2 0 %~ 0 35 % ,产物质量分数为 95 %~ 97% ,提取周期为 48h ;乙酸乙酯 -水体系选择性好 ,省去了除单宁步骤 ,使操作大为简化 ,提取率为 0 .30 %~ 0 .6 0 % ,产物质量分数为 97%~ 98% ,提取周期为 30h ,具有明显的优越性。以氯仿作沉淀剂 ,消除了叶绿素的干扰 ,提高了产物纯度。

Abstract:The active components (oligomeric flavanols,OF) in grape seeds were extracted with two solvent systems of V(acetone)∶ V (water)=2∶3 and V (ethyl acetate)∶ V (water)=8 5∶1 5 separately.The results showed that the extraction process by acetone/water system was complicated and tedious (48 h),and the yields was 0.20%~0.35% with 95%~97% purity,while the extraction process by ethyl acetate/water system was simple and rapid (30 h),obviating the procedure of tannin removal.Having higher yield (0.40%~0.60%) and purity(97%~98%),the use of ethyl acetate/water system was obviously advantageous.In the present method,the interference of chlorophyll was eliminated by using chloroform as a precipitant,so that extracts of higher purity could be obtained.
