Abstract:
The strains accumulating PHB(poly-β-hydroxybutyrate) in cells were isolated from the soil samples collected at Shenlongjia.The PHB production capacities and growth curves of three of the strains were analyzed.As results shown,PHB accumulation of strain SNJD-3-Ⅰ and strain SNJD-3-Ⅱ had reached the maximum on the sixth day and declined significantly on the seventh day.However,PHB accumulation of strain SNJD-3-Ⅲ increased continuously after declining in the growth curve.The strain SNJD-3-Ⅰ was tested as gram-positive bacteria by Gram stain,bacilli,with spore,and preliminary identified as Bacillus.The fermentation conditions for SNJD-3-Ⅰ PHB were optimized by orthogonal experiment.The experiment results showed that the optimum fermentation conditions were as follows:basic fermentation medium,glucose as carbon source,peptone as nitrogen source,carbon-nitrogen ratio of 2∶1,pH value of 7.0,32 ℃ and six days of shaking cultivation.On above conditions,the production of PHB was 14.72 g·L-1.
