摘    要:采用聚乙二醇(PEG)/盐双水相体系萃取分离米曲霉中性蛋白酶。探讨了不同成相盐及其浓度、成相聚合物PEG及其浓度、pH和NaCl浓度对中性蛋白酶萃取的影响。结果表明,适宜的双水相萃取体系为20%PEG1000/25%(NH4)2SO4(m/m)、pH8.0、不添加NaCl,此条件下中性蛋白酶上相酶活回收率和纯化倍数分别为99.88%和2.32。

Abstract:Extraction and purification of the neutral protease of Aspergillus oryzae was carried out by the polyethylene glycol(PEG)-salt aqueous two-phase systems.The effect of types and concentrations of phasing salt,the molecular weight and concentrations of phasing PEG,pH and the concentrations of NaCl on purification of the enzyme were studied.The results showed that the optimum conditions for extraction and purification of the enzyme in the system of the extraction were 20% PEG1000,25%(NH4)2SO4(m/m),but without addition of NaCl,pH8.0,resulting in recovery and the purification fold of the neutral protease in top phase could reach 99.88% and 2.32 respectively.
