摘    要:以非诺贝特酸和异丙醇为原料,采用一水合硫酸氢钠作催化剂合成了非诺贝特。通过正交试验法考察了反应时间、醇酸摩尔比、催化剂用量、带水剂和催化剂重复使用等因素对非诺贝特的酯化产率的影响。确定最佳反应条件为:以0.1mol非诺贝特酸为基准,异丙醇与非诺贝特酸物质的量之比为3.5∶1,催化剂一水合硫酸氢钠用量3.5g,带水剂二甲苯40mL,反应时间3h。目标化合物的结构和纯度经熔点测定、红外光谱、核磁共振谱、高效液相色谱等确证。改进发现一水合硫酸氢钠是一种不腐蚀设备、对环境友好的优良催化剂,酯化率可达88.6%,适合工业化生产。

Abstract:Through the esterification reaction of fenofibrate acid and isopropyl alcohol, fenofibrate was synthesized with sodium bisulfate monohydrate as catalyst. The influencing factors of the reaction, such as reaction time, molar ratio of acid and alcohol, catalyst's dosage, water-carrying reagent and reusing of catalyst were investigated. The optimal conditions were obtained as follows: basing on 0. 1 mol fenofibrate acid, the molar ratio of isopropyl alcohol to fenofibrate acid was 3. 5:1; the weight of the catalyst was 3.5 g;the dosage of water-carrying reagent(toluene)was 40 mL; the reaction time was 3.0 h. The product was identified by melting point test, IR, 1H NMR and HPLC. The results showed that sodium bisulfate monohydrate was a good catalyst for the synthesis of fenofibrate, which would not cauterize the equipment, was friendly to the environment, and could be reused. The yield of fenofibrate was 88.6%.
