磁性壳聚糖微球固定化脂肪酶研究

摘    要:以磁性壳聚糖微球为载体,通过戊二醛交联进行脂肪酶固定化,对影响脂肪酶固定化各种因素进行考察,确定最佳条件,并比较游离酶与固定化酶pH和热稳定性。结果表明,固定化适宜条件为:脂肪酶加入量5.0 mg/100 mg载体、温度40℃、时间5 h、pH 8.04、戊二醛浓度10%、最高固载率可达90.56%,酶活4034 U/g载体;与游离酶相比,固定化酶pH和热稳定性都有较宽适用范围。

Abstract:Lipase(EC3.1.1.3) was immobilized on magnetic chitosan microspheres by using glutaraldehyde as coupling reagent.Different factors influencing the immobilization were investigated,and the optimum conditions were ascertained.Comparative studies of pH and thermal stability between free lipase and immobilized lipase were also conducted;The experimental results showed that the optimal conditions for preparation immobilized lipase were lipase 5.0mg/100mg carrier,immobilization time 5 h,temperature 40℃,pH=8.04,concentration of glutaraldehyde 10%.The maximum binding efficiency reached 90.08%,and the activity of immobilized lipase was 4034.74 U/g·carrier.Therefore,the immobilized lipase holds excellent adaptability in a wider pH region and thermal stability comparable to free lipase.
