摘    要:采用Plackett-Burman试验设计和正交试验设计对影响固定化纤维素酶活的因素进行了筛选和优化.探讨了纤维素酶、壳聚糖、乙酸、乙醇、戊二醛、氢氧化钠等试剂对固定化效果的影响,筛选出制备固定化纤维素酶过程中的主要影响因素.结果表明:壳聚糖、乙酸、乙醇浓度、酶浓度分别为15g/L、2.0%、40%、100%时,以壳聚糖为载体制备固定化纤维素酶的活力最高,成型效果最好.

Abstract:Screening and optimization of key factors for cellulase immobilization on chitosan bead was studied by Plackett-Burman design and the orthogonal experiment.The main factor was determined by study on effects of chitosan,acetic acid,ethanol,sodium hydroxide,glutaraldehyde and cellulase on the activity of cellulose immobilized on chitosan.The result showed that the highest enzyme activity and best mechanic strength of immobilized cellulose occurred when the concentration of chitosan,acetic acid and ethanol are...
