RGD三肽液相合成的研究

摘    要:由N-保护的精氨酸,C-保护的甘氨酸采用混合酸酐法缩合成RG二肽,再与天冬氨酸在液相中合成RGD三肽。探讨了成酐和碱性试剂,反应温度,甘氨酸和天冬氨酸羧基保护等对缩合反应的影响。TLC跟踪反应与纯化全过程。以精氨酸计RGD三肽收率为61.4%,HPLC确认其纯度为88.66%。

Abstract:The RG Dipeptides were condensated from N-protected Arg and C-protected Gly by mixed anhydrides method. Then Asp and RG Dipeptides were used synthesize RGD Tripeptides in liquid phase. The influencing factors of condensation reaction were investigated such as anhydrides, base agent, temperature, carboxyl group protected of Gly and Asp. TLC method follows the track of reaction and purification. The yield of RGD was 61.4% for Arg. The purity was 88.66% by HPLC.
