摘    要:采用反溶剂法合成Zein-EGCG复合颗粒,用于制备负载番茄红素的Pickering乳液。根据二者复合比例和pH筛选出粒径较小的粒子进行表征,以乳析指数为指标制备不同颗粒浓度和油水比的Pickering乳液,研究剪切时间、转速以及番茄红素的浓度对负载Pickering乳液包埋率的影响,采用响应面进行优化实验,将其置于4℃和25℃测定其保留率。结果表明当复合比例为1∶0.2,pH为5时粒径大小为136.4±0.9nm;颗粒浓度为2.5%,油水比5∶5时乳析指数为2.0%±0.7%;利用响应面优化得到最佳实验条件为剪切时间为4min、剪切转速转速为12400r/min及番茄红素浓度为1.5mg/mL,负载番茄红素Pickering乳液平均包埋率为90.53%±0.63%,将其分别置于4℃和25℃条件下保存,4℃下28d保留率为57.62%±0.49%,25℃下21d保留率51.89%±0.33%。证明通过反溶剂法制备负载番茄红素Pickering乳液可提高番茄红素保留率,为后续研究与开发提供技术支持。

Abstract:Zein(Zein)-epigallocatechin gallate(EGCG)composite particles were synthesized by anti solvent method and used to prepare Pickering emulsion loaded with lycopene.The particles with smaller particle size were selected for characterization according to the two compound ratio and pH value.The Creaming Index was used to evaluate the Pickering emulsion that prepared with different granule density and oil to water rate.Study the effect of shearing lasting,rotation velocity and concentration of lycopene on the embedding rate of the burdened emulsion.Response surface was used to optimize the experiment,and its retention rate was measured at 4℃and 25℃.The results showed that the average particle diameter was just 136.4±0.9nm,when the composite ratio was 1∶0.2 and pH was 5.The Creaming Index was approximately 2.0%±0.7%when the oil-water rate was 5∶5 and the concentration of particle was 2.5%.Using response surface optimization,the best experimental conditions were obtained as follows:the shear time was 4min,the shear speed was 12400r/min and the lycopene concentration was 1.5mg/mL.The average embedding rate of the loaded lycopene Pickering emulsion was 90.53%±0.63%.It was stored at 4℃and 25℃respectively,the retention rates were 57.62%±0.49%at 4℃for 28 days and 51.89%±0.33%at 25℃for 21 days.The research showed that the lycopene Pickering emulsion prepared by anti-solvent method can improve the retention rate of lycopene,and provide technical support for further research and development。
