甲型肝炎减毒活疫苗(H2株)和灭活疫苗(L8株)诱导小鼠体液及细胞免疫应答

目的评估甲型肝炎减毒活疫苗和灭活疫苗的免疫学效果。方法用甲型肝炎减毒活疫苗(H2株)和灭活疫苗(L8株)分别经腹腔免疫BALBc小鼠后,用ELISA法检测血清中抗HAVIgG、TNFα、IFNγ和IL2;用流式细胞仪对全血中T细胞亚群分类;3HTdR掺入法检测T淋巴细胞增殖。结果减毒活疫苗组在初免后诱导的体液和细胞免疫水平均低于灭活疫苗组,加强免疫后,与灭活疫苗相当。灭活疫苗组IFNγ的峰值出现时间较减毒活疫苗组晚,但水平较高。结论两种疫苗均可诱导体液和细胞免疫应答,减毒活疫苗进行加强免疫后可诱导更好的系统免疫应答。

Humoral and Cellular Immune Responses Induced by Live Attenuated(H2) and Inactivated(L8) Hepatitis A Vaccines in Mice

Objective To evaluate the systematic immunological effect of live attenuated and inactivated hepatitis A(HA) vaccines.Methods Immunize i.p. BALB/c mice with live attenuated HA vaccine prepared with H2 strain and inactivated HA vaccine prepared with L8 strain respectively and detect the anti-HAV IgG,TNF-α,IFN-γ and IL-2 levels in sera by ELISA,the subgroup of T lymphocyte in whole blood by flow cytometer,and T lymphocyte proliferation by ~ 3H-TdR incorporation.Results After primary immunization,both the humoral and cellular immune response levels induced by live attenuated HA vaccine were significantly lower than those by inactivated HA vaccine.However,after booster immunization,both the immune response levels induced by live attenuated HA vaccine were equivalent to those by inactivated vaccine.Compared with that induced by live attenuated HA vaccine,the peak value of IFN-γ level induced by inactivated HA vaccine appeared late but was high.Conclusion Both live attenuated and inactivated HA vaccines induced humoral and cellular immune responses.The systemic immune response levels induced by live attenuated HA vaccine in booster immunization was significantly higher than that in primary immunization.
