黑曲霉TJ-1降解赤霉酸的条件优化及其特性研究

摘    要:研究了黑曲霉TJ-1对赤霉酸的降解作用,应用Plackett-Burman试验设计确定了影响其降解赤霉酸的主要因素,利用响应面分析法优化了其降解条件,探讨了黑曲霉TJ-1降解赤霉酸的特性。结果表明,黑曲霉TJ-1能够降解赤霉酸;影响其对赤霉酸降解的主要因素包括培养温度、基质中硫酸铵及硫酸镁浓度;采用优化的降解条件,该菌株对赤霉酸的降解率可达到56.90%,与所建立响应面模型的预测值(57.46%)吻合;培养体系中赤霉酸含量、菌体生物量及pH随时间的变化呈现出一定的规律性。

Abstract:The degradation of gibberellic acid (GA3) by the strain Aspergillus niger TJ-1 was studied. Plackett-Burman Design was used to screen important factors affecting its degradation, and response surface analysis was applied to optimize its degradation condition. The properties of GA3 degradation by the strain were also investigated. The results showed that Aspergillus niger TJ-1 could degrade GA3. Incubation temperature and the concentrations of ammonium sulfate and magnesium sulfate in the system were the major factors influencing GA3 degradation. The degradation rate of GA3 was up to 56.90% under the optimized condition, which was close to the predictive value from response surface model (57.64%). A regular change of GA3 contents, biomass and pH in the system with incubation time was observed.
