96孔板高通量选育产乙醇酵母

摘    要:以巴斯德毕赤酵母(Pichia pastoris),安琪酵母(Angel yeast),酿酒酵母(Sacchro-myces cerevisiae)等4株菌为出发菌株,经过紫外诱变,96孔板高通量筛选出60株,复筛选出4株.最大乙醇产量提高到76.6g/L,糖醇转化率达到了0.488g/g.对上述4株菌进行耐乙醇性能驯化,发酵耐乙醇能力达到15%.

Abstract:From these yeast trains,which can be easy attained in own laboratory,60 strains were screened through UV mutation using 96-well plates high throughput screening(HTS).4 strains were selected after advanced screening.The maximal ethanol production was enhanced to 76.6g/L and the transform rate was 0.488g/g.Through domestication with ethanol addition before fermentation,the ethanol tolerance of these 4 strains was all above 8%.The tolerance before fermentation got to 12%,the final ethanol content in fermentation broth was 15%.
