摘    要:采用单细胞Raman-D2O同位素标记技术确定了5种原油嗜蜡菌中的优势菌种;考察了其最佳生长条件,并对其代谢产物及其对原油的除蜡降黏性能进行测定。结果表明:优势原油嗜蜡菌种为SS1,属于铜绿假单胞菌属(Pseudomonas aeruginosa);其最佳生长条件为温度36 ℃、pH值7、NaCl质量浓度5 g/L;该菌种在生长代谢过程中产生糖脂类生物表面活性剂,乳化系数高达63.75%;用该菌种对原油处理120 h后,原油蜡降解率为53.16%,析蜡点及析蜡高峰点均有所降低;在36 ℃下,其对原油降黏率达到72.84%。

Abstract:Single-cell Raman-D2O isotope labeling technique was used to screen out the dominant species from five paraffin-degrading bacteria. The optimal growth conditions of strain, and their metabolites and capability to remove wax and reduce viscosity in crude oil were investigated. The results show that the dominant strain in crude oil paraffin-degrading bacteria is SS1, which belongs to Pseudomonas aeruginosa. The optimal growth conditions are the temperature of 36 ℃, the pH value of 7 and NaCl mass concentration of 5 g/L. The strain produces glycolipid type biosurfactants, in the process of growth and metabolism, with the emulsification index of 63.75%. Crude oil was treated with strain SS1 for 120 h, and the rate of wax degradation is 53.16%, the wax appearance point and the wax appearance peak point both decrease, and the rate of viscosity reduction reaches 72.84% at 36 ℃.
