丁醇高产菌株诱变育种及发酵条件优化研究

摘    要:利用低温等离子体和亚硝基胍(NTG)对兼性厌氧产丁醇芽孢杆菌(Bacillus sp.)C2菌株进行复合诱变,分离筛选得到3株丁醇和总溶剂产量均有明显提高的突变菌株,其发酵7%玉米醪液后丁醇和总溶剂产量分别达到12.59g/L、12.44g/L、12.74g/L和20.36g/L、20.14g/L、20.79g/L,比出发菌株分别提高21.6%~24.5%和15.4%~19.1%.对编号为414的突变菌株发酵条件进行优化,在100mL三角瓶发酵体系中,该菌株的最适发酵条件为装液量100mL,种龄24h,接种量10%,pH 7(自然),37℃静置发酵72h,在此条件下,414菌株发酵7%玉米醪液产丁醇和总溶剂量分别达到12.58g/L~13.77g/L和21.25g/L~22.27g/L.

Abstract:Bacillus sp.C2,a facultative anaerobic butanol producing strain,was compound with low temperature plasma and nitrosoguanidine(NTG).Three high yield mutant strains were and their butanol and total solvent yields form 7% corn mash reached 12.59g/L,12.44g/L,12.74g/L and 20.36g/L,20.14g/L,20.79g/L,respectively,which was 21.6%~24.5% and 15.4%~19.1% higher than the original strain.The optimal fermentation conditions of the mutant strain 414 was as following(in 100mL flask fermentation system): 100mL liquid volume...
