摘    要:建立超高效液相色谱法测定肉类食品中甲氧氯普胺含量的检测方法。样品用乙腈提取,经HLB小柱净 化后,采用C18色谱柱(2.1 mm×100 mm,1.7 μm),乙腈-0.1%甲酸水溶液(20∶80,V/V)为流动相,流速 0.3 mL/min,检测波长275 nm,柱温35 ℃,进样量3.0 μL。结果表明:甲氧氯普胺在1~100 mg/L范围内呈良好的线 性关系,R2=0.999 5,检出限为0.25 mg/kg,定量限为0.5 mg/kg,相对标准偏差为0.79%~5.27%(n=12),平均回 收率为87.0%~106.0%。所建立的方法适用于检测肉类食品中甲氧氯普胺的含量。

Abstract:An analytical method for the quantitative determination of metoclopramide in meat by ultra-high performance liquid chromatography(UPLC) was established in this paper. Samples were extracted with acetonitrile and then the extract was purified using an HLB cartridge. The chromatographic separation was accomplished on a C18 column(2.1 mm × 100 mm, 1.7 μm)using a mobile phase consisting of acetonitrile and 0.1% formic acid in water(20:80, V/V) at a flow rate of 0.3 mL/min. The detection wavelength was 275 nm and the column temperature was 35 ℃. The injection volume was3.0 μL. The results showed that the linear range of metoclopramide was from 1 to 100 mg/L(R^2 = 0.999 5). The limit of detection was 0.25 mg/kg, and the limit of quantitation was 0.5 mg/kg. The precision, expressed as relative standard derivation(RSD) was 0.79%–5.27%(n = 12). The average recovery was 87.0%–106.0%. The developed method proved to be applicable to the determination of metoclopramide in meat.
