纤维素酶的滤纸酶活和CMC酶活的测定

摘    要:采用3,5-二硝基水杨酸(DNS)为显色剂、滤纸或CMC为底物,测定纤维素酶的滤纸酶活(FPA)和CMC酶活(CMCA)。分析确定了酶活测定用波长为530nm,参比溶液应为失活酶、底物和DNS等共热的反应物,比较了两种底物的酶活测定方法,结果表明:纤维素酶的CMC酶活比滤纸酶活高,酶对水溶性底物有较高的活力,吸附对酶的活性部位与纤维素分子链段的结合及催化均有很大影响。

Abstract:With 3,5-dinitrosalicylic acid (DNS) as developer and filter paper or CMC as the substrate, filter paper enzyme activity (FPA) and CMC enzyme activity (CMCA) of cellulase were determined. Through analysis, wave length for determining enzyme activity was set as 530nm. The reference solution should be the reactant of inactivated enzyme. substrate and DNS. The determination methods for two kinds of substrates were compared. The result indicated that CMCA of cellulase is higher than FPA and enzyme is more active to water solable substrate. adsorption has great influence on the bonding of active part of cellulase with molecular chain of cellulose and the catalysis.
