摘    要:目的研究连续传代的KMB-17细胞株的遗传稳定性。方法将第23代KMB-17细胞连续传代至60代,按常规染色体分析方法,观察不同代次细胞的染色体数目和结构变化。选取核基因组和线粒体基因组中高突变的15个STR位点,经PCR扩增和电泳分析微卫星不稳定性。结果KMB-17细胞株经连续传代培养至56代仍未发生染色体数目和结构改变,微卫星位点也未发生突变。结论KMB-17细胞株连续传代培养后,仍能保持细胞生物学特征及遗传特征的稳定,40代以前的细胞作为疫苗生产基质是安全的。

Genetic Stability of KMB-17 Cell Strain
Abstract:Objective To study the genetic stability of KMB-17 cell strain during continuous subculture.Methods The KMB-17 cell strain passage 23 was subcultured continuously to passage 60,and the number and structure of chromosome of various passages were observed.Thirteen highly mutant STR sites from nucleic genome and two highly mutant STR sites from mitochondrial genome were selected for analysis of microsatellite instability(MSI)by PCR and electrophoresis.Results After subculture to passage 56,neither number nor structure of chromosome of KMB-17 cell strain showed any change,and no mutations at microsatellite sites were observed.Conclusion The KMB-17 cell strain showed stable cellular biological and genetic characters after continuous subculture.The strain within 40 passages was safe as a matrix for vaccine production.
