| 摘 要： | 介绍了一种新的ＹＡＣ ＤＮＡ的回收方法。该方法先用脉冲电泳分离并回收ＹＡＣ条带，然后连接引物用ＰＣＲ扩增ＹＡＣ ＤＮＡ。这一方法回收的ＹＡＣ ＤＮＡ一是量多，节省制备探针的时间；二是产物总的染色体跨度大，有利于染色体原位杂交，经ＦＩＳＨ鉴定证明该方法得到的ＹＡＣ ＤＮＡ对ＹＡＣ的定位和嵌合的鉴定非常有效。

| Abstract: | A new method of prefering FISH probes from YAC DNA has been developed. The main procedures ofthe method are as follows: (1) YAC DNA is segregated and recovered with paulse field gel electrophoresis;(2)DNA is digested with Alu I and ligated to a linker, and then amplified with linker primer by PCR.Using this method, the yield and the representation of YAC DNA are suitable for using as probe of FISH. |
