摘    要:介绍了一种新的YAC DNA的回收方法。该方法先用脉冲电泳分离并回收YAC条带,然后连接引物用PCR扩增YAC DNA。这一方法回收的YAC DNA一是量多,节省制备探针的时间;二是产物总的染色体跨度大,有利于染色体原位杂交,经FISH鉴定证明该方法得到的YAC DNA对YAC的定位和嵌合的鉴定非常有效。

Abstract:A new method of prefering FISH probes from YAC DNA has been developed. The main procedures ofthe method are as follows: (1) YAC DNA is segregated and recovered with paulse field gel electrophoresis;(2)DNA is digested with Alu I and ligated to a linker, and then amplified with linker primer by PCR.Using this method, the yield and the representation of YAC DNA are suitable for using as probe of FISH.
