新型脂质体吸附乙肝疫苗的稳定性及其抗病毒作用

目的 研究新型脂质体及其与乙肝疫苗结合物的稳定性以及结合物抗病毒作用。方法 通过外观观察、粒径检测及诱导BALB/c小鼠产生特异性抗体IgG1和IgG2a的水平考察稳定性;免疫HBV转基因小鼠,检测其血清中HBsAg和抗-HBs的滴度。结果 4℃存放1年,脂质体有少量沉淀产生,粒径变大;而脂质体与乙肝疫苗结合物全部沉淀,粒径变大,但均易摇匀,无结块;结合物4℃存放1年,诱导BALB/c小鼠产生特异性抗体IgG1和IgG2a的水平没有下降;HBV转基因小鼠于免疫后2、3、4个月,体内的HBsAg的A值较免疫前有显著下降。抗-HBs在初次免疫2个月后的阳转率为100％。结论 脂质体吸附乙肝疫苗稳定性好,且有一定的抗病毒作用。

Stability and Antivirus Effect of A Novel Liposome-HBsAg Conjugant

Objective To study the stabilities of a novel liposome and its conjugant to HBsAg, as well as the antivirus effect of the conjugant. Methods Observe the appearance and measure the sizes of the liposome and its conjugant to HBsAg. Detect the specific IgG1 and IgG2a levels in BALB/c mice immunized with the conjugant. Inoculate the conjugant into mice transformed with HBV gene and detect the titers of HBsAg and anti-HBs in sera.Results After storage at 4℃ for one year,a small quantity of preapitate formed in the liposome, and the sizes of particles were larger than those before storage. However, the conjugant was completely precipitated,and the sizes of particles became larger. Both the precipitates of liposome and conjugant were easy to be shaken up.The specific IgG1 and IgG2a levels in mice immunized with the conjugant after storage at 4℃ for one year showed no decrease. However, compared with those before inoculation, the A values of HBsAg in HBV transgenic mice 2,3 and 4 months after inoculation were significantly decreased.The positive conversion rate of anti-HBs 2 months after the first inoculation were 100% .Conclusion The liposome-HBsAg conjugant showed good stability and definite antivirus effect.
