丙酮-超声法提取叶绿素a工艺的优化

摘    要:利用正交实验法对丙酮-超声法提取、分光光度法测定叶绿素a工艺的4个操作条件(超声功率、超声时间、离心转速、离心时间)进行优化。实验材料使用了实验室培养的小球藻,避免了藻类种类对于测定结果的影响结果表明:样品在225W功率下超声30min,6000r/min转速下离心9min,搅拌后相同条件下,重复超声、离心步骤,可以得到较好的提取效果。且环保高效,方便大批量样品的测定。

Abstract:The four operating conditions of acetone-ultrasouno extracuon ano ~pec-ut,-u determination of chlorophyll a process (ultrasonic power, ultrasonic time, rotational speed, centrifugation time) were optimized by orthogonal experiment. The experimental material is chlorella brought up in laboratory to avoid the impact of the algae The results show that the sample sonicated at 225W power 30min, 6000r/min centrifuged 9min, and that after stirring and repeating ultrasonic and centrifugation step, it can get a better extraction effect.
