米曲霉HDF-7蛋白酶的分离纯化及酶学性质研究

摘    要:对米曲霉 HDF-7 所产蛋白酶进行分离纯化,经硫酸铵沉淀和凝胶过滤层析得到电泳纯的蛋白酶,经SDS-PAGE 电泳测定其相对分子质量约为30 KDa.该酶的最适反应温度为50℃,最适作用pH值为7.0,该酶在20℃时具有良好的热稳定性,在pH6.0~8.0的条件下酶是相对稳定的,Mn2+对该蛋白酶有明显的激活作用,Cu2+对该蛋白酶有强烈的抑制作用,Km值为78μg/mL,最大反应速度V max为6.29μg/min.

Abstract:The protease produced by Aspergillus oryzae HDF-7 was purified by using ammonium sulfate precipitations and gel filtration chromatography.The purified protease showed on SDS-PAGE as a single band with an apparent molecular weight of 30 KDa.The optimum temperature was 50℃ and the optimum pH was 7.0;It was found that the protease was stable at 20 ℃ and at pH 6.0-8.0;Protease activity was strongly activated by Mn2+ and strongly inhibited by Cu2+.The Km values of the protease for casein at 40 ℃ was 78 μg /mL with Vmax 6.29 μg/min.
