黑皮鸡枞菌多糖超声辅助提取条件优化及抗氧化活性研究

摘    要:目的 响应面法优化超声波提取法提取黑皮鸡枞菌(Oudemansiella raphanipes)多糖的工艺。方法 以黑皮鸡枞菌多糖的提取率为指标,采用超声波提取法从黑皮鸡枞菌中提取多糖,以超声功率、超声提取时间和碱浓度作为单因素变量,利用单因素试验结合响应面法优化确定超声波提取黑皮鸡枞菌多糖的最佳提取工艺。采用乙醇分级法黑皮鸡枞菌多糖进行乙醇分级,并对分级多糖的抗氧化活性进行研究。结果 结果表明优化得到的黑皮鸡枞菌多糖最佳工艺条件为:超声功率151.8 W、超声时间102.0 min和碱浓度0.05 mol/L,在此条件下,黑皮鸡枞菌多糖提取率最高,达到15.40% ± 0.20%,与响应面预测值16.02%相近。此外,5种乙醇分级多糖均具有一定的抗氧化活性。结论 响应面模型是成功的、可行的,80%以上分级多糖的羟基自由基清除率和还原力最强,80%分级多糖的DPPH自由基清除能力最强。

Abstract:Objective To optimize the extraction process conditions of polysaccharides from Oudemansiella raphanipes by ultrasonic-assisted extraction method, and explore the antioxidant activity of the polysaccharides. Methods Taking the extraction rate of polysaccharide as evaluation indicators, the polysaccharides from Oudemansiella raphanipes were obtained using ultrasonic-assisted extraction method. The ultrasonic power, ultrasonic time, and solvent concentration were selected as single factor variables. Based on single factor experiment, the extraction process conditions of Oudemansiella raphanipes polysaccharides were optimized by response surface method. The Oudemansiella raphanipes polysaccharides were fractionated and obtained using different concentration alcohol, and the antioxidant activities of the different fractionated polysaccharides were compared. Results The optimal extraction conditions were as follows: Ultrasonic power 151.8 W, ultrasonic time 102.0 min, and solvent concentration 0.05 mol/L. Under the optimal extraction conditions, the extraction yield of Oudemansiella raphanipes polysaccharide was the highest, reaching 15.40%±0.20%, which was close to the predicted value of 16.02%. The influence of single factor on extraction yield of Oudemansiella raphanipes polysaccharide was as follows: Ultrasonic power, ultrasonic time, and solvent concentration. The 5 kinds of ethanol graded polysaccharides had different degrees of antioxidant activities. Conclusion The applied response surface method model is successful and feasible, the hydroxyl radicals scavenging rates and reducing power of 80% above are the strongest, and the antioxidant activity 80% graded polysaccharide is the strongest among them.
