RP-HPLC测定烟用香精香料中的绿原酸及其异构体

摘    要:建立了反相高效液相色谱法(RP-HPLC)法测定烟用香精香料中的绿原酸及其2种异构体(新绿原酸、隐绿原酸)的方法,并采用该方法对9个香精香料样品中的绿原酸及其异构体进行检测。结果表明:①用甲醇振荡提取5 min,即可萃取溶解出其中的绿原酸及其异构体;②用Symmetry C18色谱柱,乙腈和0.1%甲酸的水溶液为流动相梯度洗脱,325 nm处进行检测,20 min内目标物达到完全分离;③该方法在不同浓度范围内的回收率为:新绿原酸101.3%~104.5%,绿原酸99.9%~102.7%,隐绿原酸100.0%~104.4%。6次重复实验的RSD范围为1.8%~4.7%;④咖啡酊和云烟浸膏中有较高含量的绿原酸及其异构体,不同来源的咖啡酊中绿原酸异构体的含量有所差别,其他部分样品中检测出了绿原酸。

Abstract:A method of reversed phase high performance liquid chromatography (RP HPLC) with VWD was developed for the determination of 3 isomers of chlorogenic acid(neochlorogenic acid, chlorogenic acid, cryptochlorogenic acid)in tobacco flavor- ings. Using this method, the contents of 3 chlorogenic acid isomers in 9 tobacco flavoring samples were quantitatively deter- mined respectively. It is showed that: @3 chlorogenic acid isomers in tobacco flavorings were extracted with methanol by oscil- lation of 5 min; @Separation within 20 min by RP HPLC was achieved by gradient elution on a Symmetry C18 column with ace- tonitrile and water(0. 1 % formic acid) as mobile phase at detection wavelength 325 nm; @The R. S. D of 6 repeat tests were in the range of 1.8% - 4.7% with the recoveries range 101.3% - 104.5%for neoehlorogenic acid, 99.9% - 102.7% for chloro- genic acid, 100.0% - 104.4% for cryptochlorogenic acid; (4)High content of chlorogenic acid isomers were presented in coffee tincture and Yunyan tobacco concrete; The content of chlorogenic acid isomers was different according to the source of coffee tincture. Chlorogenic acid was detected in part of the samples.
