酶辅助超声提取阿尔泰金莲花总黄酮工艺研究

摘    要:阿尔泰金莲花是预防感冒的常用药材,黄酮类化合物是其主要的化学成分。为了深入开发这一药食兼用资源,以黄酮提取率为考察指标,通过单因素试验考察酶种类、酶添加量、料液比、pH、提取时间和提取温度对阿尔泰金莲花总黄酮提取率率的影响,进一步采用Box-Behnken中心组合试验及响应面分析,优化阿尔泰金莲花总黄酮的提取工艺。结果表明,纤维素酶最适合辅助超声提取阿尔泰金莲花总黄酮,最佳工艺条件为:pH为4,料液比1:40,超声功率90W,酶添加量量:1.4%,乙醇浓度47.0%,提取时间38.0min,提取温度39.0℃;在此条件下,黄酮提取率达到了18.0%以上。该工艺简单高效,可用于阿尔泰金莲花总黄酮的提取。

Abstract:In order to improve the extraction of total flavonoids from Trollius altaicus,the extraction rate of total flavonoids was selected as the evaluation index,effects of enzyme type,pH value,enzyme addition amount,solidliquid ratio,extraction time,extraction temperature and ultrasonic frequency on the extraction rate of flavonoids were investigated by single factor experiment.The response surface methodology was further carried out to optimize the process for enzymatic-assisted ultrasonic extraction of total flavonoids from T.altaicus.Results showed that cellulase is the the most suitable enzyme for the assisted ultrasonic extraction of total flavonoids from T.altaicus,and the optimal process were as follows:pH=4,the ratio of solid to liquid 1:40(g/mL),ultrasonic power 90 W,the amount of enzyme 1.4%,ethanol concentration 47.0%,enzymolysis temperature 39.0℃,and extraction time 38.0 min;the extraction rate was more than 18.0% under these conditions.This technique is simple and efficient,and can be used to extract flavonoids from T.altaicus.
