结晶紫与蛋白质相互作用的电化学行为的研究

摘    要:结晶紫(CV)是一种三苯甲烷类染料,常用作生物染色剂和无机离子的显色剂,在pH4.5的NaAc-HAc缓冲液中结晶紫与蛋白质能够相互作用形成复合物,使结晶紫在-0.82 V(vsSCE)处的极谱还原峰峰电流下降,在最佳条件下,峰电流的下降值同牛血清白蛋白(BSA)的浓度在0.5~35mg/L范围内呈线性关系,其线性回归方程为Δip=38.7 728.84CBSA,相关系数r=0.998(n=10),检出限为0.2 mg/L。可将该方法应用于血清样品的测定,结果满意。

Abstract:Crystal violet can react with bovine serum albumin to produce an electrochemically non-active supermolecular crystal violet-BSA in the buffer solution of NaAc-HAc(pH4.5).Crystal violet has well-defined first order derivative linear-sweep polarographic wave at-0.82 V(vs SCE).Adding BSA into the crystal violet solution results in decrease of peak current without any change of peak potential.The decrease of peak current is proportional to BSA concentration from 0.5 to 35 mg/L.The limit of detection is 0.2 mg/L,and the correlation coefficient is 0.998.This method can be applied to determine BSA concentration.
