AOAC方法检测大豆异黄酮的浅析

摘    要:对AOAC方法检测大豆异黄酮进行了分析。从提取温度、提取时间、甲醇体积分数对大豆异黄酮检测的影响,以及提取后样品的大豆异黄酮含量的日间稳定性等方面进行了分析。结果显示:最佳提取温度为65℃,最佳提取时间为2.0h,最佳甲醇体积分数为80%。该方法在5d内日间精密度良好,稳定性高。AOAC方法中6种大豆异黄酮标准品线性关系良好,可靠性高,只采用6种大豆异黄酮异构体便可检测出大豆异黄酮含量。AOAC方法操作简单,成本较低,对国内大豆异黄酮的测定具有较好的参考意义。

Preliminary Analysis of AOAC Official Method for Determination of Soybean Isoflavones

Abstract:The AOAC official method for determination of soybean isoflavones is analyzed.The effects of extraction temperature,extraction time,methanol volume fraction on the determination of soybean isoflavones are investigated and the day to day precision of the results is discussed as well.The results show that the optimum extraction temperature is 65 ℃C,the optimum extraction time is 2.0 h and the optimum methanol volume fraction is 80%,and this method has good day to day precision.Under this method and HPLC conditions,the six standard isoflavones are well separated and the standard curves show good linear relationship.In this method only six kinds of soybean isoflavone standard samples are used but the soybean isoflavones content in soybean and soybean products can be well detected.The AOAC official method possesses the feature of low cost,simple operation,reliable and stable data,which has important guiding significance in the detection of soybean isoflavones at home.
